摘要
目的研究益心泰有效组分对慢性心力衰竭兔心肌组织TGF-β mRNA及蛋白表达的影响。方法采用阿霉素耳缘静脉注射+丙基硫氧嘧啶灌胃制作兔慢性心衰模型。比较各组兔心率、尿量、E峰与A峰的比值(E/A)、心肌组织转化生长因子-β(TGF-β) mRNA及蛋白表达等。结果益心泰有效组分干预后慢性心衰兔心率、尿量情况改善,差异有统计学意义(P <0.01),E/A比值升高,差异有统计学意义(P <0.01),心肌组织TGF-β mRNA及蛋白表达下降,差异有统计学意义(P <0.01)。结论益心泰有效组分可提高心功能,改善心衰,其机制可能与抑制心肌组织TGF-β mRNA及蛋白表达有关,且中、高剂量组疗效优于低剂量组。
Objective To study the effects of the active componets of Yixintai on the protein and mRNA expression of TGF-β in myocardial tissue of chronic heart failure(CHF) rabbits. Methods CHF rabbits model is established by intraperitioneally injected with adriamycin and propylthiouracil suspension. The heart rate, urine volume, the ratio of E/A, the protein and mRNA expression of TGF-β in myocardial tissue are observed. Results After intervention of the active componets of Yixintai, the heart rate and urine volume of CHF rabbits are improved, the difference is statistically significant(P < 0.01), the ratio of E/A is increased, the difference is statistically significant(P < 0.01), the protein and mRNA expression of TGF-β are decreased, the difference is statistically significant(P < 0.01). Conclusion The active componets of Yixintai can improve heart function, improve heart failure. The mechanism may be that it can inhibit the protein and mRNA expression of TGF-β in myocardial tissue. Especially, the medium and high dose groups are more effective.
