复方鱼腥草合剂对db/db小鼠心肌损伤的影响 被引量:4

摘要 目的探讨复方鱼腥草合剂(鱼腥草、牛蒡子)对糖尿病小鼠心肌超微结构的影响。方法选取8只db/m小鼠为正常组;48只db/db小鼠分为模型组、盐酸二甲双胍组、复方鱼腥草合剂水提物组、醇提物组、挥发油组及完整成分组,每组8只、给药8周,记录小鼠体重及血糖,8周后Elisa试剂盒法检测血清胰岛素、转化生长因子(TGF-β_1)、纤连蛋白(FN)、胰高血糖素样肽-1(GLP-1)水平,取心肌组织采用透射电镜法观察小鼠心肌组织超微结构的变化。结果与模型组比较,醇提物、挥发油、完整成分组血清TGF-β_1,FN水平降低(P<0.05),GLP-1含量升高(P<0.05)。心肌超微结构中肌丝排列相对整齐,线粒体肿胀程度小,改善明显,水提物组改善不明显,且以完整成分组改变最为显著。结论复方鱼腥草合剂对糖尿病所致心肌损伤有一定改善作用,其保护机制可能与下调TGF-β_1、FN水平,减少心脏胶原纤维沉积,诱导GLP-1分泌有关。 Objective To investigate the effects of different extracts from compound Herba Houttuyniae on development of diabetic cardiomyopathy in db / db mice. Methods db / m mice were in the normal group,and db / db mice were divided into six groups according to their treatment ways randomly: model group,metformin hydrochloride group,water extract group,ethanol extract group,volatile oil group,and mixture group( mixture of above three extract) of compound herba houttuyniae(n = 8 for each). The change of body weight and fasting blood glucose( FBG) of mice were detected. Fins,transforming growth factor-beta 1( TGF-β1),fibronectin( FN),glucagon-like peptide-1( GLP-1) were measured by ELISA after treatment with 8 weeks. The structures of myocardial ultrastructure were observed under electron microscope. Results Compared with normal group,the serum level of TGF-β1,FN in ethanol extract group,volatile oil group,and mixture group were decreased(P〈0. 05). The level of GLP-1 in treatment groups except metformin hydrochloride group were increased(P〈0. 05). And the myocardial ultrastructure were improved especially in mixture group. Conclusion The compound herba houttuynia can lessen the cardiac pathologicalchange,and the mechanism may related to the decrease of the level of TGF-β1,FN and the increase of GLP-1.
