摘要
目的探讨归芪方对糖尿病(DM)大鼠心肌组织转化生长因子-β1(TGF-β1)mRNA表达的影响。方法SD大鼠腹腔注射链脲佐菌素(STZ)建立DM模型,给予归芪方或贝那普利灌胃治疗。8周后测定各组血糖、血脂、心脏重量、体重的改变,荧光定量PCR法检测心肌TGF-β1mRNA的表达。结果归芪方治疗组大鼠血糖、血脂异常明显改善;心肌TGF-β1mRNA表达,归芪方优于贝那普利组(P<0.01)。结论归芪方可调节糖尿病大鼠受损的糖、脂代谢异常,逆转心肌肥厚,抑制心肌TGF-β1mRNA的表达,改善心功能,延缓糖尿病心血管并发症的进展。
Objective To investigate the effect of Guiqi Mixture on the expression of transforming growth factor-β1 (TGF-β1) mRNA in myocardium of diabetic rats. Methods The rat model of diabetic cardiomyopathy was established by single intraperitoneal injection of 60 mg/kg streptozotocin (STZ) . Then animals were treated with Guiqi Mixture or benazepril. After treatment for 8 weeks, blood glucose,lipid, heart weight and body weight were measured. The expression of TGF-β1 mRNA in myocardium was determined by fluorescence quantitative PCR. Resuits Guiqi Mixture lowered the blood glucose and lipid in diabetic rats. The expression of TGF-β1 mRNA in cardiac tissues of Guiqi Mixture group was decreased compared with that in benazepril group (P〈 0.01). Conclusion Guiqi Mixture may alleviate the disorder of glucose and lipid metabolism as well as slow down the progress of cardiovascular complications in diabetic rats by inhibiting the expression of TGF-β1 mRNA.
