摘要
目的探究结肠癌患者病理组织及血清中IL-8、VGEF及MMP-7水平与肿瘤分期的相关性。方法选取我院收治的90例经病理确诊的结肠癌患者作为研究对象,设为观察组。选取同期30例健康体检者为对照组。采用酶联免疫吸附(ELISA)法检测患者血清IL-8、VEGF和MMP-7水平;免疫组化SP法测定患者结肠癌组织中IL-8、VEGF和MMP-7阳性表达,分析其与肿瘤分期关系;同时采用Pearson相关性分析评估结肠癌组织、血清IL-8与VGEF、MMP-7之间的相关性。结果观察组血清IL-8、VEGF及MMP-7分别为(351.75±44.18)ku/L、(511.42±32.62)ng/L和(35.84±3.74)μg/L,均显著高于健康人群(P均<0.05);且结肠癌患者Dukes分期越重,血清IL-8、VEGF及MMP-7水平越高(P<0.05)。结肠癌组织中IL-8、VEGF及MMP-7阳性表达率显著高于癌旁正常组织(P<0.05)。Pearson相关性分析发现,患者血清IL-8与VEGF、MMP-7显著正相关(r=0.88,P<0.05;r=77.00,P<0.05);结肠癌组织中IL-8也与VEGF、MMP-7显著正相关(r=0.66,P<0.05;r=47.00,P<0.05)。结论IL-8、VEGF及MMP-7在结肠癌组织和血清中均高表达,与肿瘤Dukes分期密切相关;同时结肠癌组织及血清中IL-8表达与VEGF及MMP-7明显正相关,可作为临床诊疗及预后的重要评估指标。
Objective To explore the correlation between IL-8,VGEF and MMP-7 levels of the pathological tissues and serum and tumor stage in colon cancer patients.Methods 90 cases of colon cancer diagnosed by pathology admitted to our hospital were selected as the study objects,and were all set as the observation group.A total of 30 healthy cases were selected as the control group at the same period.Serum IL-8,VEGF and MMP-7 levels of patients in the two groups were detected by enzyme-linked immunosorbent assay(ELISA).Meanwhile,the expression of IL-8,VEGF and MMP-7 levels in colon cancer tissues of the observation group were detected by immunohistochemical SP method to analyze the relationship between IL-8,VEGF,MMP-7 levels and tumor staging.At the same time,Pearson correlation analysis was used to evaluate the correlation of IL-8 and VGEF,MMP-7 between in colon cancer tissue and serum.Results Serum IL-8,VEGF and MMP-7 in the observation group were(351.75±44.18)ku/L,(511.42±32.62)ng/L and(35.84±3.74)μg/L,respectively,which were significantly higher than that of healthy people(P<0.05).With the increase of Dukes staging of colon cancer patients,serum IL-8,VEGF and MMP-7 were increased(P<0.05).The positive expression rate of IL-8,VEGF and MMP-7 in colon cancer tissue were significantly higher than that of normal tissue adjacent to carcinoma(P<0.05).Pearson correlation analysis found that the IL-8 contents in colon cancer tissue and serum were significant positive correlated with VEGF and MMP-7(P<0.05).Conclusion IL-8,VEGF and MMP-7 were both highly expressed in colon cancer tissue and serum,which was closely related to the Dukes staging of tumor.IL-8 expression in the colon cancer tissue and serum presented significantly positive correlation with VEGF and MMP-7,which could be an important evaluation index for clinical diagnosis,treatment and prognosis.
