原花青素复合物的降血脂作用及其含量测定 被引量:4

摘要 目的评价葡萄皮渣中活性物质原花青素(OPC)、白藜芦醇(Res)和葡萄籽油(GSO)提取物对于降血脂效果的交互作用并测定各主要成分的准确剂量。方法建立高血脂小鼠模型,采用2×2×2析因实验,将不同配比的复合物给予小鼠30 d,测定小鼠血清总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白胆固醇(LDL-C)和高密度脂蛋白胆固醇(HDL-C)水平,利用高效液相色谱(HPLC)法确定了复合物中OPC和Res对小鼠降血脂作用的准确剂量。结果分别用200、50、20 mg·kg^(-1)OPC、Res和GSO组成的复合物降脂效果最优。HPLC法测得OPC、Res含量分别为26.5%(RSD=0.59%)、23.8%(RSD=0.29%)。结论OPC、Res和GSO复合物具有协同降脂效果,HPLC法测定了复合物中主要成分的含量,对于葡萄酒产业的可持续发展具有重要意义。 Objective In order to evaluate the interaction of oligomeric proantho cyanidins(OPC),resveratrols(Res)and grape seed oil(GSO)in grape residue on hypolipidemic,and to determine the exact dosage of the main ingredient.Methods A 2×2×2 factorial experiment based on murine hyperlipemia model was established in this study.Different combinations of oligomeric proantho cyanidins complex were given to Kunming mice for 30 days,then the levels of serum total cholesterol(TC),triglyceride(TG),low-density lipoprotein cholesterol(LDL-C)and high-density lipoprotein cholesterol(HDL-C)were measured.The accurate dose of OPC and Res in the complex were determined by HPLC method.Results The optimal combination of the complex was OPC 200 mg·kg^(-1),Res 50 mg·kg^(-1)and GSO 20 mg·kg^(-1).And the main component content results showed that the complex contained 26.5%(RSD=0.59%)of OPC and 23.8%(RSD=0.29%)of Res.Conclusion The complex of OPC,Res and GSO had a synergistic effect on lowering blood lipids.The HPLC method could effectively determine the content of the main components of the complex.This study has great significance for the sustainable development of the wine industry.
