摘要 目的:对猪胰岛素样生长因子结合蛋白(IGFBP3)基因的结构与功能进行生物信息学分析。方法:用分子生物学软件分析猪IGFBP3基因序列和IGFBP3蛋白的理化性质、二级结构、结构域、信号肽、跨膜结构、磷酸化和糖基化位点,以及不同动物IGFBP3氨基酸序列的同源性和遗传进化。结果:猪IGFBP3基因编码区长882个碱基对(bp),编码293个氨基酸。猪IGFBP3蛋白分子式为C_(1 359)H_(2 182)N_(418)O_(411)S_(24),相对分子质量为31 722.27,理论等电点为8.97,不稳定系数为48.17,脂肪系数为61.06,平均亲水系数为-0.592。IGFBP3蛋白存在1个长27个氨基酸残基的信号肽,但无跨膜结构,二级结构以无规卷曲(65.87%)和α螺旋(20.82%)为主,含1个胰岛素生长因子结合蛋白同源物(IB)结构域和1个甲状腺球蛋白Ⅰ型重复(TY)结构域,有26个磷酸化位点(包括17个丝氨酸位点、5个苏氨酸位点和4个酪氨酸位点)和3个N-糖基化位点。序列同源性方面猪IGFBP3蛋白与水牛、黄牛、山羊、绵羊、猫、狗、小鼠、大鼠、鸡和鸭的氨基酸序列同源性分别为91.2%、90.8%、87.8%、87.8%、87.2%、86.9%、82.3%、81.2%、73.9%和73.3%。系统进化树构建显示,猪IGFBP3蛋白与水牛、黄牛、绵羊和山羊的亲缘关系最近。结论:预测结果为进一步研究猪IGFBP3基因及其编码蛋白的结构和生物学功能提供基础。 Objective:The bioinformatics analysis of pig IGFBP3(insulin-like growth factor binding proteins 3)gene was conducted.Methods:The biological softwares were used to analyze the sequence of IGFBP3 gene and the physicochemical properties,secondary structure,domain,signal peptide,transmembrane structure,phosphorylation and glycosylation sites of IGFBP3 protein in pig,as well as the homology and genetic evolution of IGFBP3 amino acid sequences in different animals.Results:The coding region of pig IGFBP3 gene was 882 bp and encoded 293 amino acids.The molecular formula of pig IGFBP3 was C _(1359) H _(2182) N _(418) O _(411) S _(24),the molecular weight was 31722.27,the theoretical isoelectric point was 8.97,the instability index was 48.17,the aliphatic index was 61.06,and the grand average of hydropathicity was-0.592.There was a signal peptide with 27 amino acid residues,but no transmembrane structure in IGFBP3 protein.The secondary structure was mainly composed of random coil(65.87%)and alpha helix(20.82%).It contained an IB(insulin growth factor-binding protein homologues)domain and a TY(thyroglobulin type I repeats)domain.There were 26 phosphorylation sites(including 17 serine sites,5 threonine sites,4 tyrosine sites)and 3 N-glycosylation sites.The amino acid sequence homology of IGFBP3 protein between pig and water buffalo,cattle,goat,sheep,cat,dog,mouse,rat,chicken and duck were 91.2%,90.8%,87.8%,87.8%,87.2%,86.9%,82.3%,81.2%,73.9%and 73.3%,respectively.The phylogenetic tree construction showed that the pig IGFBP3 protein was the closest relaed to water buffalo,cattle,sheep and goat.Conclusion:The predicted results provided the basis for further studying the structure and biological function of pig IGFBP3 gene and its encoded protein.
