摘要
目的:对鸡胰高血糖素受体(GCGR)基因的结构与功能进行生物信息学分析。方法:应用生物信息学软件解析鸡GCGR基因序列及其蛋白的理化性质、蛋白结构、结构域、信号肽、跨膜结构和亚细胞定位等特性,以及不同动物GCGR氨基酸序列的同源性和遗传进化程度。结果:鸡GCGR基因编码区长1491 bp,编码氨基酸496个,预测理论等电点和分子量分别为8.43和56.69201 kD,是一种较稳定的疏水性蛋白;存在4个N-糖基化位点和45个磷酸化位点,1个7tmB1_GCGR结构域;二级结构主要为α-螺旋,与三级结构预测结果一致;同源性分析发现,该基因编码的蛋白氨基酸序列与鹌鹑的相似性最高,其次为疣鼻天鹅、棕榈尾鸭、仓鸮等禽类。亚细胞定位分析发现,该基因主要定位在细胞质膜上。结论:该结果为进一步研究鸡GCGR基因及其编码蛋白的结构和生物学功能提供基础。
Objective:The structure and function of glucagon receptor(GCGR)gene were analyzed by bioinformatics.Methods:Bioinformatics software was used to analyze the physicochemical properties,protein structure,domain,signal peptide,transmembrane structure and subcellular localization of chicken GCGR gene sequence and its protein,as well as the homology and genetic evolution degree of GCGR amino acid sequence in different animals.Results:The coding region of chicken GCGR gene is 1491 bp,encoding 496 amino acids.The predicted theoretical pI and molecular weight are 8.43 and 56.69201 kD,respectively,indicating that it is a stable hydrophobic protein.There were 4 N-glycosylation sites,45 phosphorylation sites and a 7tmB1_GCGR domain.The secondary structure is mainlyα-helix,which is consistent with the predicted results of tertiary structure.The sequence of amino acid of the mute protein encoded by the mute gene was the most similar to that of quail,followed by mute swan,palm tail duck and barn owl.Subcellular localization analysis showed that the gene mainly located in the plasma membrane of the cell.Conclusion:The predicted results will provide a basis for further study on the structure and biological functions of chicken GCGR gene and its encoded protein.
