摘要
目的:用不同浓度的脂多糖(LPS)对大鼠进行宫腔灌注,尝试构建更符合子宫内膜损伤条件的大鼠子内膜损伤动物模型,并对该模型进行评价。方法:选取SD大鼠24只,将其分为LPS 10 mg/mL组、LPS 20 mg/mL组(此二组合称为LPS处理组大鼠)和对照组。分别向LPS 10 mg/mL组、LPS 20 mg/mL组和对照组大鼠的宫腔内注射10 mg/mL的LPS、20 mg/mL的LPS、同等体积的生理盐水。干预48 h后处死三组大鼠,切除其子宫,并对子宫进行苏木精-伊红(HE)染色及免疫组化染色,观察其子宫内膜的形态学改变。结果:造模后,与对照组大鼠相比,LPS处理组大鼠的子宫内膜层明显变薄,宫腔腔隙增大,子宫内膜上皮细胞排列紊乱,间质充血水肿,腺体数量明显减少,且LPS 20 mg/mL组大鼠的子宫内膜层变薄尤为明显。造模后,LPS处理组大鼠子宫内膜和肌层中肿瘤坏死因子-α(TNF-α)的表达水平显著高于对照组大鼠,且随着LPS诱导剂量的加大,大鼠子宫内膜和肌层中TNF-α的表达水平逐渐升高。结论:LPS可有效地诱导大鼠子宫内膜损伤,LPS的最佳诱导剂量为20 mg/mL。LPS可作为一种损伤诱导剂成功构建大鼠子宫内膜损伤模型。
Objective:Different concentrations of lipopolysaccharide(LPS)were used to intrauterine perfusion in rats,to try to establish a rat model of endometrial injury that is more suitable for the conditions of endometrial injury,and to evaluate the model.METHODS:Twenty-four SD rats were selected and divided into LPS10 mg/mL group,LPS 20 mg/mL group(these two combinations were called LPS treated rats)and control group.Rats in the LPS 10 mg/mL group,the LPS 20 mg/mL group and the control group were intrauterine injected with 10 mg/mL LPS,20 mg/mL LPS and the same volume of normal saline.After 48 h of interven tion,the three groups of rats were sacrificed,and their uterus was removed,and the uterus was stained with hematoxylin-eosin(HE)and immunohistochemical staining,and the morphological changes of endometrium were observed.Results:After model building,compared with the control group,the endometrium in the LPS group was significantly thinned,the uterine lacuna was enlarged,the endometrium epithelial cells were disordered,the interstitial hyperemia and edema were observed,and the number of glands was significantly reduced,especially in the LPS 20 mg/mL group.After modeling,the expression level of tumor necrosis factor-α(TNF-α)in endometrium and musculum of rats in LPS treatment group was significantly higher than that in control group,and the expression level of TNF-αin endometrium and musculum of rats in LPS treatment group increased gradually with the increase of LPS induced dose.Conclusion:LPS can effectively induce endometrial injury in rats,and the optimal dose of LPS is 20 mg/mL.LPS can be used as an inducer to construct a rat endometrial in jury model.
