营养性肥胖高血压大鼠动物模型的建立与评价

摘要 目的采用喂养高脂饲料的方法建立一种贴近临床肥胖高血压的疾病动物模型并进行评价。方法120只5周龄雄性自发性高血压大鼠(SHR)随机分为普通饲料组(SHR-ND组,n=12)及高脂饲料组(SHR-HF组,n=108),以12只周龄/性别相匹配的正常血压京都种大鼠(WKY)做对照并喂养普通饲料(WKY-ND组,n=12)。造模干预10周后,将SHR-HF组中体质量位于上游前1/3的大鼠判定为肥胖高血压大鼠(OBH-HF组,n=36)。造模前及造模后每2周测量体质量、体长、腹围、收缩压水平并计算Lee指数,评估造模前后易激惹程度并积分,检测造模后的三酰甘油(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、空腹血糖(FBG)、空腹胰岛素(FIN)并计算胰岛素抵抗指数(HOMA-IR)。结果造模前,WKY-ND组、SHR-ND组及SHR-HF组3组间体质量、Lee指数、腹围无统计学差异(P>0.05);SHR-ND组及SHR-HF组2组间收缩压水平无统计学差异(P>0.05),但二者均明显高于WKY-ND组(P<0.01);WKY-ND组、SHR-ND组及SHR-HF组3组间易激惹程度积分无统计学差异(P>0.05)。造模后,OBH-HF组体质量明显高于SHR-ND组(P<0.01);OBH-HF组Lee指数、腹围明显大于WKY-ND组及SHRND组(P<0.01);OBH-HF组收缩压水平明显高于WKY-ND组及SHR-ND组(P<0.01);OBH-HF组易激惹程度积分明显高于WKY-ND组及SHR-ND组(P<0.01);OBH-HF组TC、LDL-C水平高于SHR-ND组(P<0.05),HDL-C水平明显低于SHR-ND组(P<0.01)。结论以高脂饲料喂养自发性高血压大鼠可诱导建立肥胖高血压疾病动物模型,并在行为学、理化指标等方面与临床肥胖高血压基本相符。 Objective To establish and evaluate a disease animal model close to clinical obesity and hypertension by feeding highfat diet.Methods A total of 1205-week-old male spontaneously hypertensive rats(SHR)were randomly divided into a normal diet group(SHR-ND group,n=12)and a high-fat diet group(SHR-HF group,n=108).Another 12 age-/sex-matched Wistar-Kyoto rats(WKY)with normal blood pressure were used as control and fed with normal diet(WKY-ND group,n=12).After 10 weeks of modeling intervention,the rats whose body weight was in the top 1/3 in the SHR-HF group were judged to be obese hypertensive rats(OBH-HF group,n=36).Before modeling and every 2 weeks after modeling,the body mass,body length,abdominal circumference and systolic blood pressure were measured,the Lee index was calculated,the irritability degree before and after modeling was evaluated and integrated,and the triglyceride(TG),total cholesterol(TC),low density lipoprotein cholesterol(LDL-C),high density lipoprotein cholesterol(HDL-C),fasting blood glucose(FBG),fasting insulin(FIN)were detected and insulin resistance index(HOMA-IR)were calculated after modeling.Results Before modeling,there was no statistically significant difference in the body mass,Lee index,and abdominal circumference among the WKY-ND group,SHR-ND group and SHR-HF group(P>0.05).There was no statistically significant difference in the systolic blood pressure between the SHR-ND group and SHR-HF group(P>0.05),but they both were significantly higher than the WKY-ND group(P<0.01).There was no statistically significant difference in the irritability score among the WKY-ND group,SHR-ND group and SHR-HF group(P>0.05).After modeling,the body mass of the OBH-HF group was significantly higher than that of the SHR-ND group(P<0.01).The Lee index and abdominal circumference of the OBH-HF group were significantly greater than those of the WKY-ND group and the SHR-ND group(P<0.01).The systolic blood pressure level of the OBH-HF group was significantly higher than that of the WKY-ND group and SHR-ND group(P
