摘要
为筛选出对禽沙门菌具有显著拮抗效应的益生菌,从健康肉鸡消化道分离菌株,经细菌形态观察、生化试验鉴定及16S rDNA基因序列分析,共鉴定出14株唾液乳杆菌。体外琼脂扩散试验结果显示,WLS001、WLS002、WLS003、WLS004这4株分离株分别对鼠伤寒沙门菌、肠炎沙门菌、鸡白痢沙门菌、鸡伤寒沙门菌具有明显抑制作用。pH及胆盐耐受性试验结果表明,WLS001耐受性最强,在pH为2.0、胆盐浓度0.2%条件下均能生存。药敏结果显示,4株菌均对万古霉素耐药。安全性试验表明,所有唾液乳杆菌均没有溶血环,试验组雏鸡体重较对照组增重显著(P<0.05)。组织病理切片显示,脾、肝、盲肠及十二指肠形态均正常。结果表明,4株鸡源唾液乳杆菌具有良好的生物学特性和安全性,具备进一步开发微生态制剂的潜力。
In order to screen probiotics with significant antagonistic effect on Salmonella in poultry,a total of 14 Lactobacillus salivarius strains were identified from the digestive tract of healthy broilers by morphological observation,biochemical identification and 16S rDNA gene sequencing. The results showed that the four isolated strains WLS001,WLS002,WLS003 and WLS004 had obvious inhibitory effect on Salmonella typhimurium,Salmonella enteritidis,Salmonella pullorum and Salmonella gallinarum respectively by agar diffusion test.The results of pH and bile salt tolerance tests showed that WLS001 had the best tolerance and could survive at pH2.0 and bile salt concentration of 0.2%. The four strains were resistant to vancomycin by antibiotic sensitity.The security and animal test results showed that four Lactobacillus salivarius strains had no hemolytic ring.Compared with the negative control,the weight of chickens in the experimental group was increased(P<0.05),and there was no significant difference in the morphology of spleen,liver,cecum and duodenum. Therefore,the four Lacto-bacillus salivarius strains isolated from chickens have good biological characteristics and probiotic safety,which can be used as a potential probiotic for further study.
