IL-10和IL-17基因单核苷酸多态性与感染性角膜炎严重程度的相关性及预后评估价值

Correlation between IL-10 and IL-17 gene single nucleotide polymorphisms and the severity of infectious keratitis and its prognostic value

摘要 目的探讨白细胞介素-10(IL-10)和IL-17基因单核苷酸多态性与感染性角膜炎疾病严重程度的相关性及预后评估价值。方法选择庆阳市人民医院眼科2017年5月-2019年12月收治的感染性角膜炎患者92例作为研究组,将研究组患者分为轻度组31例,中度组33例,重度组28例,选择同期医院收治的角膜损伤但未发生感染的患者50例作为对照组。采用聚合酶链式扩增反应(PCR)及一代测序法对IL-17F基因rs2397084位点、IL-10基因rs1800871位点多态性进行检测,采用酶联免疫吸附法(ELISA)检测血清中IL-10、IL-17水平。结果两组IL-17F基因rs2397084位点、IL-10基因rs1800871位点基因型分布均符合Hardy-Weinberg平衡;两组IL-17 F基因rs2397084位点、IL-10基因rs1800871位点基因型、等位基因分布差异均有统计学意义(P<0.05);研究组IL-17 F基因rs2397084位点GG基因型、IL-10基因rs1800871位点TT基因型频率高于对照组(P<0.05);校正年龄、性别等因素后,携带IL-17F基因rs2397084位点G等位基因、IL-10基因rs1800871位点T等位基因发病风险分别增加0.568、1.017倍;研究组不同病变严重程度IL-17F基因rs2397084位点、IL-10基因rs1800871位点等位基因分布差异均无统计学意义;预后不佳组患者血清IL-10、IL-17水平高于预后佳组患者(P<0.05)。结论IL-17F基因rs2397084位点、IL-10基因rs1800871位点多态性与感染性角膜炎发病风险增加有关,但与疾病严重程度无关,其血清水平对于判断预后具有一定评估价值。 OBJECTIVE To investigate the correlation between interleukin-10(IL-10)and IL-17 gene single nucleotide polymorphisms and the severity of infectious keratitis and its prognostic value.METHODS A total of 92 patients with infectious keratitis admitted to the Department of Ophthalmology,Qingyang People′s Hospital from May 2017 to Dec.2019 were selected as the study group,and the study group was divided into 31 cases in the mild group,33 cases in the moderate group,and 28 cases in the severe group,50 patients with corneal injury without infection who were admitted to the hospital during the same period were selected as the control group.The polymorphism of IL-17 F gene rs2397084 locus and IL-10 gene rs1800871 locus were detected by polymerase chain amplification reaction(PCR)and first-generation sequencing method,and the serum IL-10 and IL-17 levels were detected by enzyme-linked immunosorbent assay(ELISA).RESULTS The genotype distribution of the two groups of IL-17 F gene rs2397084 locus and IL-10 gene rs1800871 locus were in accordance with Hardy-Weinberg equilibrium.The distribution differences of IL-17 F gene rs2397084 locus,IL-10 gene rs1800871 locus genotype,allele of two groups were significant(P<0.05).The frequency of GG genotype at rs2397084 site of IL-17 F gene and TT genotype at rs1800871 site of IL-10 gene in the study group were significantly higher than those in the control group(P<0.05).After adjusting for age,gender and other factors,the risk of the G allele at the rs2397084 locus of the IL-17 F gene and the T allele at the rs1800871 locus of the IL-10 gene increased by 1.568 and 2.017 times,respectively.The study group had different severity of the disease and the IL-17 F gene There was no significant difference in the distribution of rs2397084 locus of IL-17 F gene and rs1800871 locus of IL-10 gene alleles in the study group with different disease severity.The serum IL-10 and IL-17 levels in the poor prognosis group were significantly higher than those in the good prognosis group(P<0.05).CONCLUSION
