摘要
目的探讨白细胞介素-17(IL-17)不同位点基因多态性与复发性流产(RSA)易感性的关联。方法选取2015年01月-2019年12月期间到我院就诊RSA患者180例设为RSA组;同时选取同期产检健康早孕孕妇180例设为对照组。采用酶联免疫吸附法(ELISA)检测血清中IL-17浓度;应用采用PCR-SSCP法检测IL-17A和IL-17F的基因型,并经Hardy-weinberg遗传平衡定律验证;分析各位点基因型与IL-17水平关系;回归分析IL-17基因型与RSA发生风险关系。结果RSA组患者IL-17浓度高于对照组(P<0.05)。两组IL-17基因rs2275913和rs763780位点基因频率均符合Hardy-weinberg遗传平衡定律。RSA组rs2275913位点的A等位基因频率高于对照组(P<0.05)RSA组rs763780位点的C等位基因频率低于对照组(P<0.05)。rs2275913和rs763780位点突变型者血清IL-17浓度高于对照组(P<0.05)。rs2275913 AG和AA基因携带者发生RSA风险的相对危险度分别为GG等位基因的2.012倍和2.545倍,rs763780 TC、TT基因携带者发生RSA风险的相对危险度分别为CC等位基因的1.735倍和1.478倍(P均<0.05)。结论RSA发生与IL-17的rs2275913、rs763780位点多态性有关,更高的A和T等位基因频率预示更高的RSA发病风险。
Objective:To investigate the association between interleukin-17(IL-17)gene polymorphism and recurrent abortion(RSA)Methods:180 cases of RSA patients diagnosed in our hospital from January 2015 to December 2019 were selected as RSA group,and at the same time,180 cases of healthy pregnant women in the same period were selected as the control group.The serum IL-17 concentration was detected by enzyme-linked immunosorbent assay(ELISA),and the genotypes of IL-17A and IL-17F were detected by PCR-SSCP,and verified by Hardy Weinberg′s law of genetic equilibrium;The relationship between IL-17 genotype and RSA risk was analyzed by regression analysis.Results:The concentration of IL-17 in RSA group was higher than that in control group(P<0.05).The gene frequencies of rs2275913 and rs763780 of IL-17 gene in the two groups were in accordance with Hardy Weinberg equilibrium law.The A allele frequency of rs2275913 in RSA group was higher than that in control group(P<0.05).The C allele frequency of rs763780 in RSA group was lower than that in control group(P<0.05).The serum IL-17 concentration of rs2275913 and rs763780 mutations was higher than that of control group(P<0.05).rs2275913 AA and AA carriers had 2.012 and 2.545 times higher risk of RSA than GG allele,and rs763780 TC and TT gene carriers had 1.735 and 1.478 times higher risk of RSA than CC allele,respectively(P<0.05).Conclusion:The occurrence of RSA was associated with rs2275913 and rs763780 polymorphisms of IL-17,higher A and T allele frequencies indicated a higher risk of RSA.
