平喘颗粒对哮喘小鼠肺组织TLR2和TLR4表达的影响 被引量:6

摘要 目的:观察平喘颗粒对哮喘小鼠肺组织Toll样受体2(toll-like receptor2,TLR2)和TLR4表达的影响。方法:将30只BALB/c小鼠随机分为对照组、模型组、平喘颗粒(31.2 g·kg^(-1))组,每组10只。除对照组外,采用腹腔注射加雾化卵清蛋白(ovalbumin,OVA)致敏液[100μg OVA+1mgAl(OH)_(3)]制备慢性哮喘小鼠模型。造模成功后给予相应的药物干预8周,ELISA法检测血清中白细胞介素-4(interleukin-4,IL-4)、IL-5和IL-13的水平;Western Blot法检测肺组织TLR2和TLR4蛋白表达水平;HE染色观察肺组织病理改变情况。结果:与模型组比较,平喘颗粒组小鼠血清中IL-4、IL-5和IL-13水平明显降低(P<0.05),肺组织TLR2和TLR4蛋白表达明显下调(P<0.05),肺组织病理损伤有明显改善。结论:平喘颗粒可缓解哮喘气道炎症,其机制可能与调控TLR2、TLR4表达有关。 Objective:To observe the effect of Pingchuan Granule on the expression of toll-like receptor2(TLR2)and TLR4 in lung tissue of asthmatic mice.Methods:30 BALB/c mice were randomly divided into control group,model group and Pingchuan Granule(31.2 g·kg^(-1))group,with 10 mice in each group.Except the control group,mouse models of chronic asthma were prepared by intraperitoneal injection plus aerosolized ovalbumin(OVA)sensitizing solution[100μg OVA+1 mg Al(OH)_(3)].After the model was successfully established,the corresponding drug intervention was given for 8 weeks,and the levels of interleukin-4(IL-4),IL-5 and IL-13 in the serum were detected by ELISA method;Western Blot method was used to detect the protein expression levels of TLR2 and TLR4 in lung tissue;HE staining was used to observe the pathological changes of lung tissue.Results:Compared with the model group,the levels of IL-4,IL-5 and IL-13 in the serum of mice in the Pingchuan Granule group were significantly reduced(P<0.05),and the expression of TLR2 and TLR4 proteins in the lung tissue was significantly down-regulated(P<0.05).The pathological damage has been significantly improved.Conclusion:Pingchuan Granules can alleviate airway inflammation in asthma,and its mechanism may be related to the regulation of the expression of TLR2 and TLR4.
