摘要
目的:优化款冬花萜类成分的提取条件,并研究其降脂作用。方法:采用四因素三水平正交试验筛选款冬花萜类成分的最优提取方法;采用高脂饲料诱导肥胖模型,小鼠随机分组为空白对照组、奥利司他阳性组、肥胖模型组、款冬花萜类成分药液低、中、高剂量组,连续灌胃给药45 d后取血、腹部脂肪及肝脏;测定血清甘油三酯、总胆固醇及高、低密度脂蛋白含量,计算Lee′s指数、脂肪系数、肝脏系数,对肝组织进行油红O染色。结果:正交试验得到最佳提取条件为:85%乙醇和1.3%盐酸,超声时间70 min,液料比20∶1(mL/g);与肥胖模型组比较,高、中、低剂量款冬花提取液均抑制了小鼠体质量增长;降低了肝脏系数与脂肪系数;降低甘油三酯、总胆固醇和低密度脂蛋白水平,升高高密度脂蛋白水平;降低肝脏脂滴含量。结论:款冬花萜类成分具有降脂作用。
Objective:To optimize the extraction conditions of terpenoids from the flower buds of Tussilago farfara,and study its lipid-reducing effect.Methods:Four factors and three levels orthogonal experiment was used to select the optimal extraction method of terpenoids from the flower buds of Tussilago farfara.The obesity model was induced by high fat diet.The mice were randomly divided into blank control group,orlistat positive control group,obesity model control group,and low,medium and high doses groups of terpenoids from the flower buds of Tussilago farfara.After 45 days of continuous intragastric administration,blood,abdominal fat and liver were collected.The serum triglyceride,total cholesterol and high-density lipoprotein,low-density lipoprotein contents were determined,The Lee′s index,adipose coefficient and liver coefficient were calculated,and liver tissue was stained with oil red O.Results:The optimal extraction conditions obtained by orthogonal experiment were as follows,85%ethanol and 1.3%hydrochloric acid,ultrasonic 70 min,liquid-solid ratio 20∶1(mL/g).Compared with the obesity model group,the high,medium and low doses of the flower buds of Tussilago farfara extract inhibited the weight growth of mice.The liver coefficient and fat coefficient were decreased.The triglyceride,total cholesterol and low-density lipoprotein level was reduced.High-density lipoprotein level content was increased,and liver lipid droplets were decreased.Conclusion:The terpenoids from the flower buds of Tussilago farfara have the effects of lipid-reducing.
