体外高糖条件下ALA/GLA抑制VEGFR-3/PI3K表达影响视网膜内皮细胞活动 被引量:1

ALA/GLA INHIBITS THE EXPRESSION OF VEGFR-3/PI3K AND AFFECTS THE ACTIVITY OF RETINAL ENDOTHELIAL CELLS UNDER HIGH GLUCOSE CONDITION IN VITRO

摘要 目的探究高糖条件下α-亚麻酸和γ-亚麻酸对视网膜内皮细胞的增殖、侵袭、凋亡和细胞周期的影响及VEGFR-3/PI3K调控作用。方法取1w龄的大鼠分离原代视网膜内皮细胞,细胞分为对照(Control)、高糖(HG)、α-亚麻酸(ALA+HG)、γ-亚麻酸(GLA+HG),α-亚麻酸+γ-亚麻酸+高糖(ALA+GLA+HG)共5组。免疫荧光鉴定细胞,CCK8检测细胞的增殖活性,Transwell检测细胞侵袭能力,流式细胞检测细胞周期和细胞凋亡率,WB检测VEGFR-3/PI3K的蛋白表达量。结果分离的内皮细胞含有血凝因子Ⅷ,鉴定该细胞是视网膜内皮细胞,高糖上调VEGFR-3和PI3K蛋白表达,ALA和GLA抑制高糖条件下VEGFR-3和PI3K蛋白表达,高糖抑制细胞的增殖活性和侵袭能力并促进了细胞凋亡,ALA和GLA可以促进高糖条件下细胞的增殖和侵袭。GLA抑制高糖条件下细胞凋亡。结论高糖条件下ALA和GLA通过抑制VEGFR-3和PI3K表达来促进细胞的增殖和侵袭,抑制视网膜血管的增生,GLA可以抑制凋亡,起到保护视网膜内皮细胞的作用。[营养学报,2021,43(1):65-70] Objective To explore the effects of α-linolenic acid and γ-linolenic acid on the proliferation, invasion, apoptosis and cell cycle and the regulation of VEGFR-3/PI3 K in the retinal endothelial cells under high glucose condition. Methods Primary retinal endothelial cells were isolatd from 1-week-old rats. The cells were divided into control(Control), high glucose(HG), α-linolenic acid(ALA+HG), γ-linolenic acid(GLA+HG), α-linolenic acid+γ-linolenic acid+high glucose(ALA+GLA+ HG). Immune fluorescence was used to identify cell types, Cell counting kit-8 was used to detect cell proliferation activity, Transwell assay was used to detect cell invasion ability, flow cytometry was used to detect cell cycle and apoptosis rate, and Western blotting was used to detect VEGFR-3/PI3 K protein expression. Results The isolated endothelial cells produced hemagglutination factor Ⅷ, and the cells were identified as retinal endothelial cells. High glucose up-regulated the expression of VEGFR-3 and PI3 K proteins, ALA and GLA inhibited VEGFR-3 and PI3K protein expression under high glucose condition. High glucose inhibited cell proliferation activity and invasive ability and promoted cell apoptosis. ALA and GLA promoted cell proliferation and invasion under high glucose condition. GLA inhibited apoptosis under high glucose condition. Conclusion Under high glucose condition, ALA and GLA promote the proliferation and invasion of the cells, and inhibit the proliferation of retinal blood vessels by inhibiting the expression of VEGFR-3 and PI3 K.
