摘要
目的建立接触甲醛和乙醛人员尿液中的代谢产物甲酸和乙酸的离子色谱测定方法。方法尿液样品经离心后,使用C18固相萃取柱萃取和0.22μm水相针式滤器过滤,用离子色谱仪进行甲酸和乙酸的定性和定量分析,优化淋洗液组成和样品前处理条件等实验参数。结果C18固相萃取柱萃取尿液中的甲酸和乙酸去除杂质效果良好,回收率高。甲酸和乙酸的色谱峰面积与其质量浓度间有良好的线性关系,相关系数(r)均>0.9998,检出限分别为0.002 mg/L和0.018 mg/L;批内相对标准偏差(RSDs)为1.02%~3.84%和1.25%~4.56%,批间相对标准偏差(RSDs)为1.14%~4.41%和1.68%~4.28%,平均回收率为97.6%~100.0%和96.2%~100.5%。用该法检测某人造板厂接触甲醛及乙醛工人的晨尿,均检出甲酸和乙酸。结论该法简便、灵敏、检出限低、干扰少,适用于人体尿液中甲酸和乙酸的测定。
Objective To establish an ion chromatographic method for the detection of formic acid and acetic acid in urine of people exposed to formaldehyde and acetaldehyde.Methods The urine samples were centrifugated and purified with C18 solid-phase extraction column and filtered by 0.22μm water phase needle filter,and detected by ion chromatography for qualitative and quantitative analysis of formic acid and acetic acid.The experimental parameters such as the compositions of eluent and the conditions of sample pretreatment were optimized.Results The extraction of formic acid and acetic acid in urine by C18 solid phase extraction column showed good impurity removal effectiveness and high recovery rate.There was a better linear relationship between the chromatographic peak area of formic acid and acetic acid and their mass concentrations.The linear correlation coefficients(value r)were both>0.9998,and the limit of detection were 0.002 mg/L and 0.018 mg/L respectively.The internal relative standard deviations(RSDs)were 1.02%-3.84%and 1.25%-4.56%.The inter-batch RSDs were 1.14%-4.41%and 1.68%-4.28%.The mean recoveries were 97.6%-100.0%and 96.2%-100.5%.The morning urine samples of workers exposed to formaldehyde and acetaldehyde in a wood-based panel factory were detected by this method,and formic acid and acetic acid were all detected out.ConclusionThe method is simple,sensitive,with low detection limit and less interference.It is suitable for the detection of formic acid and acetic acid in human urine.
