摘要
目的研究藤灸四花穴对血瘀型糖尿病周围神经病变(DPN)小鼠的血管活性因子及血红素氧合酶1(HO-1)表达的影响。方法db/db小鼠分为血瘀模型组、α-硫辛酸组、假藤灸组、藤灸3个亚组,分别给药、藤灸四花穴治疗6周,检测热痛觉阈、血糖及血清ET、NO、NOS、MDA含量,光镜观察胰腺组织形态,Weston-blot检测HO-1蛋白。结果藤灸3个亚组的热痛觉阈改善明显(P<0.05);藤灸25 min组的血糖、ET、NO、NOS、MDA改善显著(P<0.01);藤灸3个亚组胰腺组织切片HE染色发现炎症表现减轻;藤灸3个亚组的HO-1表达有所加强。结论藤灸四花穴能明显改善血瘀型DPN小鼠的周围神经功能和血瘀病理,也能减轻胰腺慢性炎症,其作用机制可能在于改善血管活性因子、抗氧化因子、及加强HO-1的表达,最终改善微循环。
Objective To study the effect of rattan moxibustion on expressions of vascular activity factor and heme oxygenase 1(HO-1)in mice with blood stasis type of diabetic peripheral neuropathy.Methods The db/db mice were divided into the blood stasis model group,the alpha lipoic acid group,sham rattan moxibustion group,three rattan moxibustion subgroups.The treatment lasted 6 weeks.The blood glucose,ET,NO,NOS,MDA and thermal pain threshold were observed and the pancreas was observed under light microscopy.Weston-blot was used to detect the HO-1 protein.Results After 6 weeks of rattan moxibustion,the threshold of heat and pain sensation and blood sugar in the 3 rattan moxibustion groups was significantly improved,ET NO,NOS,MDA was significantly improved in the three rattan moxibustion subgroups.The chronic inflammation of pancreas was reduced,and HO-1 expression was enhanced.Conclusions Rattan moxibustion at Sihua points can ameliorate the blood glucose of DPN mice with blood stasis,and can improve the blood stasis index.It has a significant effect of promoting blood circulation and removing blood stasis,reducing chronic inflammation of pancreas,and improving peripheral neural function.The mechanism is related to improving oxidative stress and enhancing HO-1 expression.
