左卡尼汀对睾丸生精功能损伤模型小鼠保护作用的研究 被引量:2

摘要 目的评估与探讨左卡尼汀(LC)治疗睾丸生精功能损伤模型小鼠的有效性和安全性。方法将40只4周龄雄性C57BL/6小鼠随机分为5组,分别为正常对照组、白消安造模后0.9%氯化钠溶液灌胃组(白消安组),以及白消安造模后低剂量LC灌胃(低LC)组、中剂量LC灌胃(中LC)组和高剂量LC灌胃(高LC)组,每组8只。采用白消安35 mg/kg腹腔内注射构建无精子症小鼠模型。建模4周后,正常对照组予正常饮食,白消安组予0.9%氯化钠溶液灌胃;低、中、高LC组分别予200、400、800 mg/(kg·d)剂量的LC连续灌胃4周。记录小鼠活动进食情况、毛发变化和体重;灌胃4周后采集血液和睾丸组织并称重,行睾丸组织石蜡包埋切片与H-E染色;记录睾丸器官指数、血睾酮(T)、黄体生成素(LH)、卵泡刺激素(FSH)水平,以及睾丸组织Johnsen评分。结果造模后,5组小鼠均存活。正常对照组、白消安组、低LC组、中LC组、高LC组小鼠的睾丸器官指数分别为(7.0±2.9)、(3.0±0.7)、(3.1±0.8)、(2.4±0.2)、(2.6±0.3)。白消安组、低LC组、中LC组、高LC组小鼠睾丸器官指数较正常对照组均显著降低(P值均<0.001)。白消安组与低LC组、中LC组和高LC组睾丸器官指数的差异无统计学意义(P值均>0.05)。正常对照组、白消安组、低LC组、中LC组和高LC组小鼠性激素水平的差异均无统计学意义(P值均>0.05)。正常对照组、白消安组、低LC组、中LC组、高LC组小鼠睾丸组织Johnsen评分分别为(9.9±0.1)、(4.3±0.3)、(4.8±0.3)、(5.4±0.4)、(5.6±0.5)分,4组造模组小鼠的睾丸组织Johnsen评分较正常对照组均显著降低(P值均<0.001),低、中、高LC组的睾丸组织Johnsen评分均显著高于白消安组(P值分别为0.018,<0.001、<0.001),中、高LC组的睾丸组织Johnsen评分较低LC组均显著升高(P值分别为0.022,<0.001),高LC组与中LC组间的差异无统计学意义(P>0.05)。与白消安组相比,低、中、高LC组生精小管排布� Objective To evaluate and discuss the safety and efficacy of L-carnitine on testicular spermatogenesis injury induced by chemotherapy in mice.Methods Forty male C57BL/6 mice of 4 weeks old were randomly divided into 5 groups(n=8):normal control group,busulfan group,low-dose L-carnitine group,moderate-dose L-carnitine group,and high-dose L-carnitine group.The azoospermia mice model was established by intraperitoneal injection of busulfan 35 mg/kg.All mice were fed with normal water and food for 4 weeks.Additionally,normal saline was given in the busulfan group;200,400 and 800 mg/(kg·d)L-carnitine were intragastrically administrated in low-,moderate-and high-dose L-carnitine groups,respectively.The food intake,hair changes and body weight of the mice were recorded.After another 4 weeks of treatment,blood and testicular tissues were collected and weighed,and blood sex hormones were measured.Paraffin embedded sections of testicular tissues were made and hematoxylin-eosin(H-E)staining were performed.Testicular organ index and Johnsen score of testicular tissue were detected.Results All the mice survived after modeling.Testicular organ index was 7.0±2.9,3.0±0.7,3.1±0.8,2.4±0.2,and 2.6±0.3 in the normal control group,busulfan group,low-,moderate-and high-dose L-carnitine groups.Testicular organ index in the normal control group was significantly higher than that in the other four groups(all P<0.001),respectively,while there was no significant difference in the testicular organ index among busulfan group,low-,moderate-and high-dose L-carnitine groups(all P>0.05).No statistical difference was found in the sex hormone levels among the five groups(all P>0.05).Johnsen score was 9.9±0.1 in the normal control group,which was significantly higher than those in the busulfan group(4.3±0.3),low-dose L-carnitine group(4.8±0.3),moderate-dose L-carnitine group(5.4±0.4),and high-dose L-carnitine group(5.6±0.5)(all P<0.001).And the Johnsen scores in the low-,moderate-and high-dose L-carnitine groups were significantly higher
