摘要
目的运用网络药理学方法研究淫羊藿改善心肌重塑的有效成分和作用机制。方法利用中药系统药理学技术平台TCMSP数据库筛选出淫羊藿口服生物利用度≥30%和类药性≥0.18的主要活性成分及其靶点基因,通过GeneCards、OMIM等数据库获取心肌重塑靶点基因。将成分靶点与疾病靶点上传至String数据库,构建中药成分靶蛋白-疾病靶蛋白互作网络,并根据其度值筛选出关键靶点。借助DAVID分析平台对关键靶标进行基因本体(GO)分析和基因组百科全书(KEGG)通路富集分析。结果共获取淫羊藿活性成分23个,与心肌重塑交集的关键靶标78个,包括白介素6(IL-6)、半胱氨酸蛋白酶(CASP)3、血管内皮生长因子A(VEGFA)、表皮生长因子受体(EGFR)、丝裂原活化蛋白激酶(MAPK)8等基因,主要富集在恶性肿瘤途径(Pathways in cancer)、肿瘤坏死因子信号通路(TNF signaling pathway)、磷脂酰肌醇3激酶/蛋白激酶B信号通路(PI3K-Akt signaling pathway)、缺氧诱导因子1信号通路(HIF-1 signaling pathway)、促乳素信号通路(Prolactin signaling pathway)等。结论本研究结果初步验证和预测了淫羊藿改善心肌重塑的作用机制,为进一步深入揭示其分子机制提供了理论基础。
Objective To use the method of network pharmacology and explore the mechanism of Yinyanghuo(Epimedii Folium)in the treatment of myocardiac remodeling.Methods The main active components and target genes of Yinyanghuo(Epimedii Folium)were collected by using TCMSP database of traditional Chinese medicine system pharmacology technology platform.The myocardiac remodeling disease target genes were obtained through GeneCards and OMIM databases.The component targets and disease targets were uploaded to String database.The interaction network of traditional Chinese medicine component target and disease target protein was constructed and the key targets were selected according to its degree.With the help of DAVID analysis platform,the key targets were analyzed by gene ontology(GO)analysis and genome encyclopedia(KEGG)pathway enrichment analysis.Results A total of 23 active components of Yinyanghuo(Epimedii Folium)were obtained,and 78 key targets of intersection with myocardiac remodeling were obtained,including IL-6,CASP3,VEGFA,EGFR,MAPK8 and other genes,which were mainly enriched in Pathway in cancer,TNF signaling pathway,PI3 K-Akt signaling pathway,HIF-1 signaling pathway,Prolactin signaling pathway and other signal pathways.Conclusion The results of this study preliminarily verified and predicted the mechanism of Yinyanghuo(Epimedii Folium)in the treatment of myocardiac remodeling,and laid a theoretical foundation for further revealing its molecular mechanism.
