摘要
目的:探讨淫羊藿素对腹膜纤维化大鼠糖原合酶激酶3β(GSK-3β)、β连环蛋白(β-catenin)的影响。方法:30只健康SD大鼠随机分为假手术组(10只)、造模组(20只)。造模组大鼠采用0.1%葡萄糖氯己定腹腔注射建立腹膜纤维化模型,造模成功后随机分为模型组和淫羊藿素组,淫羊藿素组按3 mg/kg剂量灌胃,假手术组、模型组给予等量生理盐水,连续6周,处死大鼠,取腹膜组织。采用HE染色观察腹膜组织病理变化;Masson染色观察腹膜组织胶原纤维化;免疫组化法、ELISA法检测腹膜组织GSK-3β、β-catenin水平;Western blot检测腹膜组织GSK-3β、p-GSK-3β、β-catenin蛋白表达。结果:与假手术组比较,模型组腹膜组织腹膜增厚,胶原纤维增加,腹膜组织GSK-3β、p-GSK-3β、β-catenin表达明显升高(P<0.05)。与模型组比较,淫羊藿素可减轻腹膜增厚并减少腹膜组织胶原纤维,明显降低腹膜组织GSK-3β、p-GSK-3β、β-catenin蛋白表达(P<0.05)。结论:淫羊藿素可减轻腹膜纤维化大鼠腹膜增厚及腹膜组织胶原纤维,抑制GSK-3β、β-catenin的表达,具有抗腹膜纤维化的重要作用。
Objective:To investigate the effect of icaritin on GSK-3βandβ-catenin in rats with peritoneal fibrosis.Methods:A total of 30 healthy SD rats were randomly divided into sham operation group(n=10)and model group(n=20).The rats in the model group were intraperitoneally injected with 0.1%glucose and chlorhexidine to establish the model of peritoneal fibrosis.After successful modeling,the rats were randomly divided into model group and icaritin group.Rats in the icaritin group were administered at the dose of 3 mg/kg,while the sham operation group and the model group were given the same amount of normal saline for 6 weeks.The rats were murdered and the peritoneal tissue was taken.The pathological changes of peritoneal tissue were observed by HE staining,collagen fibrosis was observed by Masson staining,and the levels of GSK-3βandβ-catenin in peritoneal tissue were detected by immunohistochemical and ELISA;Western blot was used to detect the expression of GSK-3β,p-GSK-3βandβ-catenin protein in peritoneal tissue.Results:Compared with the sham operation group,the peritoneal thickness,collagen fibers and the expression of GSK-3β,p-GSK-3βandβ-catenin in the model group were significantly higher than those in the sham operation group(P<0.05).Compared with the model group,icaritin could reduce the thickening of the peritoneum,reduce the collagen fibers in the peritoneal tissue,and significantly reduce the expression of GSK-3β,p-GSK-3βandβ-catenin protein in the peritoneal tissue(P<0.05).Conclusion:Icaritin can reduce peritoneal thickening and collagen fibers in peritoneal fibrosis rat model,and inhibit the expression of GSK-3βandβ-catenin,which may play an important role in anti-peritoneal fibrosis.
