摘要
目的探讨flotillin 2(FLOT2)在结直肠癌中的表达及其临床意义。方法采用荧光定量PCR和免疫组化方法检测4对结直肠癌组织及相应癌旁非肿瘤组织中FLOT2表达,并用免疫组化方法检测157例结直肠癌组织切片中FLOT2的表达。结合临床随访资料探讨FLOT2表达与临床病理因素及患者预后之间的关系。结果FLOT2在结直肠癌组织中的表达均高于癌旁非肿瘤组织。结直肠癌患者FLOT2表达水平与Dukes分期(P=0.013)、临床分期(P=0.013)、T分期(P=0.001)、N分期(P=0.021)、M分期(P=0.016)、组织分化(P=0.016)和生存预后(P=0.025)之间具有显著相关性,与年龄(P>0.05)、性别(P>0.05)和肿瘤位置(P>0.05)无显著相关性。Kaplan-Meier分析结果显示,FLOT2高表达的患者生存时间明显低于低表达FLOT2的患者;经log-rank检验,差异有统计学意义(P=0.013)。结论FLOT2在结直肠癌中表达上调,其差异表达与结直肠癌Dukes分期、临床分期、TNM分期、病理分级及生存预后呈显著负相关,提示FLOT2可能与结直肠癌的发生、发展相关。
Objective To study the expression of FLOT2(FLOT2)in colorectal cancer and its clinical significance.Methods Fluorescence quantitative PCR and immunohistochemistry were used to detect the expression of FLOT2 in 4 pairs of colorectal cancer tissues and corresponding non-neoplastic tissues adjacent to cancer,and to detect the expression of FLOT2 in 157 cases of colorectal cancer tissues,and explore its expression based on clinical follow-up data Correlation with clinicopathological factors and prognosis.Results The expression of FLOT2 in colorectal cancer tissues was higher than the corresponding non-neoplastic tissues adjacent to cancer.The expression intensity of FLOT2 is related to Dukes staging(P=0.013),clinical staging(P=0.013),T category(P=0.001),N category(P=0.021),M category(P=0.016),tissue differentiation(P=0.016)and survival prognosis(P=0.025)were significantly related,but not related to the patient′s age,gender,and tumor location.Kaplan-Meier analysis showed that the survival time of the FLOT2 high expression case group was significantly shorter than that of the FLOT2 low expression case group.The Log-rank test showed that the difference was statistically significant(P=0.013).Conclusion FLOT2 is up-regulated in colorectal cancer,and its expression difference is significantly negatively correlated with Dukes staging,clinical staging,TNM staging,pathological grade and patient survival prognosis,suggesting that it may be related to the occurrence and development of colorectal cancer.
