摘要
类菌孢素氨基酸(Mycosporine-like amino acids,MAAs)是一类多功能活性化合物,在大型海藻中广泛存在。以海带、裙带菜、羊栖菜、江蓠、石花菜和海萝为原料,通过一系列单因素和正交实验,建立了此6种大型海藻MAAs的适宜提取工艺,并建立了定性检测方法。结果表明,海带、裙带菜、羊栖菜、江蓠、石花菜和海萝MAAs的适宜提取温度、固液比和甲醇体积分数依次为35、30、30、30、30、25 ℃,1:60、1:60、1:60、1:60、1:60、1:50 g/mL,30%、10%、30%、10%、30%、10%。利用上述工艺条件,制备得到6种大型海藻MAAs提取物,得率分别为(39.0±3.80)%、(20.6±2.20)%、(32.8±2.60)%、(5.41±1.20)%、(9.87±0.80)%、(16.2±1.40)%。在此基础上,250~390 nm波长扫描显示6种大型海藻MAAs提取物均在330 nm附近有吸收,表明它们中存在Porphyra-334。进一步,初次确定了海带、裙带菜、羊栖菜、江蓠、石花菜和海萝中Porphyra-334含量,依次为(1.60±0.69)μg/g、(1.96±0.89)μg/g、(3.97±0.82)μg/g、(41.9±3.41)μg/g、(13.2±2.25)μg/g、(8.89±1.29)μg/g。随后,应用硅胶薄层层析,对上述6种大型海藻MAAs提取物进行检测,建立了大型海藻MAAs检测的一种简便方法,可用于大型海藻MAAs的筛选。
A series of single factor and orthogonal experiments were used to establish the extraction process of mycosporine-like amino acids (MAAs) from Laminaria japonica,Undaria pinnatifida,Hizikia fusifarme,Gracilaria arcuata,Gelidium amansii and Gloiopeltis furcata.In this process,MAAs were detected by silica gel thin layer chromatography for the first time.Namely,suitable temperature,solid-liquid ratio and methanol volume fraction of MAAs extraction were 35,30,30,30,30,25 ℃,1:60,1:60,1:60,1:60,1:60,1:50 g/mL,30%,10%,30%,10%,30%,10%,respectively.Under the above conditions,MAAs extracts from 6 kinds of marine macroalgae were prepared with the yield of (39.0±3.80)%,(20.6±2.20)%,(32.8±2.60)%,(5.41±1.20%),(9.87±0.80)% and (16.2±1.40)%,respectively.Furthermore,the results of 250~390 nm wavelength scanning showed that the MAAs extracts of six species of marine macroalgae have absorption near 330 nm.And this means that there was porphyra-334 in them.Further,the contents of porphyra-334 in Laminaria japonica,Undaria pinnatifida,Hizikia fusifarme,Gracilaria arcuata,Gelidium amansii and Gloiopeltis furcata were (1.60±0.69)μg/g,(1.96±0.89)μg/g,(3.97±0.82) μg/g,(41.9±3.41)μg/g,(13.2±2.25)μg/g and (8.89±1.29)μg/g,respectively.This was the first report on the content of porphyra-334 in six kinds of marine macroalgae.Then,silica gel thin layer chromatography was used to detect the MAAs of the six species of marine macroalgae,and a simple method for the detection of MAAs in marine macroalgae was established,which can be used for the screening of MAAs from marine macroalgae.
