Analysis of differentially expressed genes between breast cancer and normal breast tissues

摘要 [目的]基于GEO数据库中乳腺癌相关基因表达谱芯片,探讨乳腺癌组织及正常乳腺组织基因表达差异。[方法]收集2016年10月~2017年10月于医院进行乳腺癌根治术患者乳腺癌组织及其对应正常乳腺组织,共213对样本。GEO数据库提取乳腺癌相关基因表达谱芯片数据,根据筛选基因对应基因差异倍数(FC)对数值,定义表达上调和下调基因。检测关键基因mRNA表达水平,分析其水平对患者总生存期的影响。[结果]共347个基因在乳腺癌组织出现差异表达,其中包括表达上调基因207个,表达下调基因140个;乳腺癌组织与正常乳腺组织差异表达基因差异倍数前10位基因依次为KRT19、CENPE、FOXM1、CD9、BUB1B、CDC20、NDC80、CENPE、NUSAP1、GATA3;KRT19、CENPE、FOXM1高表达的乳腺癌患者总生存期显著差于低表达者(P <0.05)。[结论]基于GEO数据库中乳腺癌相关基因表达谱芯片,乳腺癌组织及正常乳腺组织存在差异表达基因,且KRT19、CENPE、FOXM1过表达为乳腺癌患者不利的预后因素,有可能成为乳腺癌基因治疗新靶点。 [Objective] To analyze the differentially expressed genes between breast cancer and normal breast tissues based on gene expression omnibus( GEO) datasets.[Method] A total of 213 pairs of breast cancer tissues and adjacent normal tissues were collected from patients underwent radical mastectomy in our hospital from October 2016 to October 2017. The chip data was downloaded from GEO database. The differentially expressed genes were screened by GEO2 R software,and divided into up-regulated and down-regulated genes according to fold change( FC). RT-PCR was used to detect the mRNA expression level of key genes in tissue samples,and the effect of mRNA expression level of key genes on the overall survival of patients was analyzed.[Result] A total of 347 differentially expressed genes were found in breast cancer tissues,including 207 up-regulated genes and 140 down-regulated genes. The top 10 differentially expressed genes between breast cancer tissue and normal breast tissue were KRT19,CENPE,FOXM1,CD9,BUB1 B,CDC20,NDC80,CENPE,NUSAP1,and GATA3 in sequence;The overall survival of breast cancer patients with highly expressed KRT19,CENPE and FOXM1 was significantly worse than that of patients with lowly expressed KRT19,CENPE and FOXM1( P < 0. 05). [Conclusion] According to the chip data downloaded from GEO database,there are differentially expressed genes in breast cancer tissues and normal breast tissues,moreover,overexpression of KRT19,CENPE,and FOXM1 are prognostic risk factors for breast cancer patients,which may become new targets for breast cancer gene therapy.
