摘要
目的探索miRNA-204-5p在乳腺癌组织和细胞中的表达,探讨miR-204-5p对乳腺癌细胞增殖、迁移、侵袭和上皮-间质转化(EMT)的影响。方法 qRT-PCR检测乳腺癌患者癌和癌旁组织,乳腺癌细胞和正常乳腺上皮细胞中miR-204-5p的表达;将miR-204-5p模拟物和阴性对照分别转染MCF-7细胞,并检测转染效率;CCK-8法检测转染对细胞增殖的影响;Transwell迁移实验和侵袭实验分别检测转染miR-204-5p后乳腺癌细胞迁移和侵袭能力的变化;Western blot检测上皮-间质转化(EMT)相关蛋白的表达。结果 miR-204-5p在乳腺癌组织表达水平显著下调,miR-204-5p在多种乳腺癌细胞株如MCF-7细胞表达量下调。miR-204-5p模拟物转染抑制MCF-7细胞增殖、迁移、细胞侵袭,E-cadhesin表达水平升高,Vimentin降低。结论过表达MiR-204-5p对乳腺癌有潜在的治疗作用。
Objective To study the expression of miRNA-204-5 p in breast cancer tissues and cells to explore the effect of miR-204-5p on breast cancer cell proliferation, migration, invasion and epithelial-mesenchymal transition(EMT). Methods RT-PCR was used to detect the expression of miR-204-5p mRNA;miR-204-5p mimics was transfected into MCF-7 cells, CCK-8 kid was used to detect the cells proliferation, Transwell was used to determine the cells migration and invasion, the expression of interstitial transformation(EMT) related proteins were detected by Western blot. Results The expression level of miR-204-5 p was significantly down-regulated in breast cancer tissues and breast cancer cell lines such as MCF-7 cells;miR-204-5p mimics inhibits the proliferation, migration and invasion of MCF-7 cells, increased E-cadhesin expression, but decreased Vimentin expression level. Conclusion MiR-204-5p has potential therapeutic effect on breast cancer.
