麻芩消咳颗粒对烟熏所致慢性支气管炎小鼠的止咳作用 被引量:2

Antitussive effect of MaQinXiaoKe granule in mice with smoke-induced chronic bronchitis

摘要 目的探讨麻芩消咳颗粒对烟熏所致慢性支气管炎模型小鼠的止咳作用及其可能机制。方法 60只ICR小鼠建立慢性支气管炎模型后随机分为模型组(B组)、肺宁颗粒3.8 g/kg给药组(C组)以及不同剂量麻芩消咳颗粒0.75、1.50、3.00 g/kg给药组(D1、D2、D3组);另设12只小鼠为对照组(A组)。记录小鼠咳嗽次数和潜伏期,采用ELISA法检测小鼠血清炎症因子含量,HE染色观察肺组织形态学变化,免疫组化检测肺组织Toll样受体4(TLR4)和NF-κB蛋白表达,Western blot法检测核因子相关因子2(Nrf2)和血红素氧合酶1(HO-1)蛋白表达。结果与B组相比,C、D1、D2、D3组小鼠咳嗽次数减少(P<0.05或P<0.01);C组和D3组小鼠咳嗽潜伏期较B组延长(P<0.05)。与A组相比,B组IL-1β和TNF-α含量以及TLR4、NF-κB、HO-1蛋白表达均增加,Nrf2蛋白表达降低(P<0.01);而C、D1、D2、D3组给药后能有效逆转上述各指标的变化过程(P<0.05或P<0.01)。结论麻芩消咳颗粒对烟熏所致慢性支气管炎模型小鼠有良好的止咳作用,其机制可能与下调TLR4/NF-κB信号通路以及上调Nrf2/HO-1信号通路有关。 Objective To investigate the antitussive effect and underlying mechanism of MaQinXiaoKe granule in mice with smoke-induced chronic bronchitis.Methods The chronic bronchitis model was established in 60 ICR mice,which then equally randomized into five groups.Another 12 mice were taken as the blank controls(group A).The mice in groups of B,C and D1,D2,D3 were pretreated with normal saline,FeiNing granule 3.8 g/kg and different concentrations of MaQinXiaoKe granule 0.75,1.50,3.00 g/kg,respectively.The attack and incubation period of cough were recorded.The contents of inflammatory factors in serum were determined by ELISA.The morphological change of lung tissues was observed by HE staining.The protein expressions of TLR4 and NF-κB in the lung tissues were measured by immunohistochemistry.The protein expressions of Nrf2 and HO-1 were detected by Western blot.Results Compared with group B,the times of cough was decreased in groups of C,D1,D2 and D3(P<0.05 or P<0.01).The incubation period of cough in groups of C and D3 was longer than that in group B(P<0.05).Compared with group A,the contents of IL-1β and TNF-α and the protein expressions of TLR4,NF-κB and HO-1 were increased,and the protein expression of Nrf2 was decreased in group B(P<0.01),while which were significantly reversed after pretreated with FeiNing granule and MaQinXiaoKe granule in groups of C,D1,D2 and D3(P<0.05 or P<0.01).Conclusion MaQinXiaoKe granule could have a good antitussive effect in mice with smoke-induced chronic bronchitis,which may be related to the downregulation of TLR4/NF-κB signaling pathway and the upregulation of Nrf2/HO-1 signaling pathway.
