摘要
目的探讨麻芩消咳颗粒的平喘抗炎作用。方法SD大鼠随机分为6组,分别为肺宁颗粒2.5 g·kg^(-1)组、麻芩消咳颗粒(1、2、4 g·kg^(-1))组、模型组和溶剂对照组。第0、7天皮下注射卵蛋白混悬液致敏。第15天开始给药并用1%的OVA溶液激发哮喘,每天给药1次,连续给药4周。给药结束后检测大鼠肺泡盥洗液中白细胞介素6(IL-6)、白细胞介素17(IL-17)含量及血清中免疫球蛋白E(IgE)、白细胞介素4(IL-4)水平,观察肺组织病理学变化,Western blot检测肺组织中PI3K、p-PI3K、AKT、p-AKT的表达水平。结果麻芩消咳颗粒2 g·kg^(-1)能够降低大鼠肺泡盥洗液中白细胞介素6、白细胞介素17含量及血清中免疫球蛋白E、白细胞介素4水平,降低PI3K、ATK磷酸化水平。结论麻芩消咳颗粒对卵蛋白诱发的大鼠过敏性哮喘模型有显著的平喘抗炎作用,与PI3K/AKT调节的炎症信号通路有关。
Objective To investigate the anti-asthmatic and anti-inflammatory effects of Maqin Xiaoke Granules.Methods SD rats were randomly divided into 6 groups:Feining Granules 2.5 g·kg^(-1)group,Maqin Xiaoke Granules(1,2,4 g·kg^(-1))group,model group and solvent control group.Rats were sensitized on day 0 and 7 by subcutaneously injection with OVA.On day 15,Rats were challenged with 1%OVA and given the drug,once a day for 4 weeks.After the administration,the levels of IL-6 and IL-17 in BALF and the levels of IgE and IL-4 in serum of the rats were measured,and the histopathologic changes of the lung tissues were examined,the expression of PI3K,p-PI3K,AKT,and p-AKT in the lung tissue were evaluated by western blot.Results Maqin Xiaoke Granules at dose of 2 g·kg^(-1)could reduce the levels of IL-6,IL-17 in the BALF and IgE,IL-4 in serum of rats,and it reduced the phosphorylation levels of PI3K and AKT.Conclusion Maqin Xiaoke Granules have significant anti-asthmatic and anti-inflammatory effects on the rat model of allergic asthma induced by egg protein,which is related to the inflammation signal pathway regulated by PI3K/AKT.
