七叶通脉胶囊对家兔全脑缺血再灌注损伤神经保护作用研究 被引量:2

Study on the neuroprotective effects of Qiye Tongmai Capsule in global cerebral ischemia-reperfusion injury rabbits

摘要 目的:探讨七叶通脉胶囊预防给药后对家兔全脑缺血再灌注模型的神经保护作用。方法:七叶通脉胶囊预防给家兔灌服10d后,夹闭双侧颈总动脉和椎动脉,并放血,使血压在40mmHg维持30min后,放开双侧颈总动脉复灌2h,制备家兔全脑缺血再灌注模型。复灌结束后取血,测定血清中LHD、Na+-K+-ATPase、GSH-Px、NSE、GFAP、SOD、MDA等因子的含量;取脑,部分测定脑组织中SOD活性和MDA含量,另一部分HE染色,观察脑组织病理结构的变化。结果:家兔全脑缺血再灌注后,与模型组比较,高剂量组能显著增强血清中GSH-Px、SOD、Na+-K+-ATPase活性(P<0.01,P<0.05),降低LDH活性、MDA含量(P<0.01),减少GFAP、NSE的释放(P<0.05,P<0.01),还能显著升高脑组织SOD活性,降低MDA含量(P<0.01),减轻脑组织的病理改变;其他组别对这些指标也有不同程度的影响。结论:七叶通脉胶囊能通过对抗氧化应激反应,抑制神经元和胶质细胞释放相关因子,减轻全脑缺血再灌注对神经的损伤。 Objective: To investigate the neuroprotective effects of Qiye Tongmai Capsule(QYTM) on rabbit model of global cerebral ischemia-reperfusion after prophylactic administration. Methods: After prevent to give QYTM for 10 d, the global cerebral ischemia-reperfusion in rabbits model were established by ligating both common carotid arteries and vertebral arteries for 30 min, in the meantime, bleeding and maintaining blood pressure at 40 mmHg. Then releasing the common carotid arteries and reperfusion for 2 h. After the end of reperfusion, collecting blood and the contents of factors of LHD, Na+-K+-ATPase, GSHPx, NSE, GFAP, SOD, MDA in blood serum were checked. Some brain was used for determining the SOD and MDA, the others were observed the pathological structure after HE staining. Results: After global cerebral ischemia-reperfusion, compared with the model, the high group of QYTM treatment could increase the activity of GSH-Px, SOD, Na+-K+-ATPase(P<0.01, P<0.05), decrease LDH and MDA(P<0.01), reduce GFAP, NSE(P<0.05, P<0.01) in blood serum, increase SOD and decrease MDA in brain tissue(P<0.01). The changes of brain tissue structure were alleviated. The other groups also could more or less influence these factors. Conclusion: The QYTM can lighten the injury of nerve caused by global cerebral ischemia-reperfusion through resisting oxidative stress and inhibiting the release of related factors of neurons and glia cells.
