摘要
目的:研究当归内酯对缺血性眩晕大鼠的保护作用及脑神经损伤修复作用。方法:除15只大鼠作为对照组外,其余75只大鼠采用右侧颈总动脉及锁骨下动脉结扎术建立缺血性眩晕大鼠模型,将造模完成后的大鼠随机分为模型组、当归内酯低剂量组、当归内酯中剂量组、当归内酯高剂量组及盐酸氟桂利嗪组,每组15只。当归内酯低、中、高剂量组根据大鼠体重分别按照20 mg/kg、40 mg/kg、80 mg/kg的当归内酯剂量给予大鼠灌胃,盐酸氟桂利嗪组给予大鼠1.5 mg/kg盐酸氟桂利嗪灌胃,对照组及模型组给予大鼠等量生理盐水灌胃,1次/d,连续给药14 d后,采用眩晕测试测定大鼠跳台逃避潜伏时间,酶联免疫吸附测试(ELISA)法测定大鼠右侧脑组织超氧化物歧化酶(SOD)活性、丙二醛(MDA)、肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)水平,苏木精-伊红(HE)染色检查大鼠右侧海马组织病理学变化,Western blotting检测大鼠右侧海马组织Bax、Bcl-2水平。结果:与模型组比较,当归内酯低、中、高剂量组及盐酸氟桂利嗪组大鼠跳台逃避潜伏时间显著缩短、右侧脑组织TNF-α、IL-1β及MDA水平显著降低(P<0.05),SOD活性显著升高(P<0.05),右侧海马组织病理学变化明显改善,Western blot结果表明,大鼠右侧海马组织Bax水平显著降低(P<0.05),Bcl-2水平显著升高(P<0.05),且各项指标变化与当归内酯的剂量表现出依赖性。结论:当归内酯能够通过缩短大鼠跳台逃避潜伏时间,降低脑组织TNF-α、IL-1β、MDA水平及海马组织Bax水平,并提高脑组织SOD活性及海马组织Bcl-2水平,改善大鼠海马组织病理学变化,进而发挥其对缺血性眩晕大鼠的保护作用。
Objective:to investigate protective effect of angelica lactone on ischemic vertigo rats and repair effect of brain nerve injury.Methods:A total of 15 rats weretaken as control group,75 rats were treated with ligation of right carotid artery and subclavian artery in order to set up rat model of ischemic vertigo.After modeling,rats were randomly divided into model group,angelica lactone low dose group,angelica lactone middle dose group,angelica lactone high dose group and flunarizine hydrochloride group,with 15 rats in each group.Angelica lactone was administered intragastrically to rats in low,middle and high does groups according to 20 mg/kg,40 mg/kg and 80 mg/kg of body weight.Flunarizine hydrochloride group was given 1.5 mg/kg flunarizine hydrochloride with gavage,while rats in the control group and model group were given the same amount of normal saline once a day.After 14 days of continuous administration,the escape latency of rats was measured by vertigo test,and the activity of superoxide dismutase(SOD)in the right brain tissue was measured by ELISA.The levels of malondialdehyde(MDA),tumor necrosis factor-α(TNF-α)and interleukin-1β(IL-1β)were measured.The histopathological changes of right hippocampus were detected by HE staining.Western blotting was used to detect the levels of Bax and Bcl-2 in right hippocampus.Results:Compared with the model group,the escape latency of Angelica lactone low,middle and high dose groups and flunarizine hydrochloride group were significantly shortened.The levels of TNF-α,IL-1βand MDA in the right brain tissue were significantly decreased(P<0.05).The activity of SOD was significantlyincreased(P<0.05).The histopathological changes of the right hippocampus were significantly improved.Western blot results showed that the level of Bax in the right hippocampal tissue was significantly decreased(P<0.05).The level of Bcl-2 was significantly increased(P<0.05),and the changes of various indexes were dependent on the dose of Angelica lactone.Conclusion:Angelica lactone can shorte
