痿证中药方抑制线粒体内质网应激减轻大鼠脊髓损伤 被引量:4

Wei Syndrome Recipe Inhibits Mitochondrial Endoplasmic Reticulum Stress and Alleviates of spinal Cord Injury Rat Model

摘要 目的研究痿证中药方对脊髓损伤(SCI)大鼠模型中氧化应激反应及内质网应激相关蛋白表达的调节作用。方法SPF级SD大鼠30只随机分为为假手术组、脊髓损伤模型组及痿证中药方组,痿证中药方组予以痿证中药方干预。术后第3~4天、1周、2周、3周、4周进行下肢运动功能评分,干预后30 d,取脊髓组织并采用HE染色法检测脊髓损伤情况,采用免疫组织化学法测定脊髓损伤大鼠内质网应激蛋白GRP78、ATF4和CHOP的含量,采用酶联免疫吸附试剂盒测定氧化应激指标MDA、CAT、SOD、GSH-Px的含量。结果术后SCI组大鼠出现了不同程度的后肢运动功能障碍,而Sham组各大鼠均未见明显运动功能损害(P<0.05)。HE染色可见Sham组中脊髓组织灰质及白质结构正常,神经元结构清晰,未见显著的脊髓受损。与Sham组比较,SCI组大鼠的脊髓组织中观察到脊髓结构严重受损,脊髓中央部的灰质和位于周围部的白质结构模糊,胞核皱缩或溶解消失并伴有不同程度的中性粒细胞浸润及大量小胶质细胞增生。治疗组脊髓结构比模型组明显改善,脊髓组织病变相对较轻,小区域的神经元坏死,少量的嗜酸性细胞增多,小胶质细胞浸润。免疫组织化学染色显示给予痿证中药方后GRP78、ATF4和CHOP的表达与SCI组相比明显减少(P<0.05),ELISA结果显示痿证中药方逆转MDA、CAT、SOD及GSH-Px活性的表达(P<0.05)。结论痿证中药方可以通过抑制线粒体损伤、减轻脊髓损伤情况、抑制氧化应激以及降低内质网应激相关蛋白的表达,从而促进脊髓损伤后大鼠运动功能的恢复。 Objective To explore the effect of Wei syndrome recipe on the regulatory role of oxidative stress response and endoplasmic reticulum stress related protein of spinal cord injury rat model.Methods 30 spf-grade SD rats were randomly divided into sham operation group,spinal cord injury model group,and the prescription group of erectile dysfunction syndrome.The prescription group of erectile dysfunction syndrome was given the prescription intervention of erectile dysfunction syndrome.Postoperative 3~4 d,1 w,2 w,3 w,4 w for lower limb motor function score,and 30 days after intervention in spinal cord tissue and HE staining method is used to detect spinal cord injury,using immune histochemical method determination of spinal cord injury in rats of endoplasmic reticulum stress protein GRP78,ATF4 and CHOP content,using enzyme-linked immunosorbent kit determination of oxidative stress indicators MDA,CAT and SOD,gsh-px levels.Results Postoperatively,rats in the SCI group presented different degrees of motor dysfunction of hind limbs,while no significant motor impairment was observed in rats in the Sham group(P<0.05).HE staining showed that the gray and white matter structures of the spinal cord tissues in the Sham group were normal,the neuronal structures were clear,and no significant spinal cord damage was observed.Compared with Sham group,the spinal cord structure of rats in SCI group was seriously damaged,the gray matter in the central part of the spinal cord and the white matter in the surrounding part were blurred,the cell nucleus was shrunkened or dissolved,and there were various degrees of neutrophil infiltration and a large number of microglial cell hyperplasia.The spinal cord structure in the treatment group was significantly improved compared with that in the model group,with relatively mild spinal cord tissue lesions,neuronal necrosis in small areas,increased number of eosinophilic cells,and infiltration of microglia.Immunohistochemical staining showed that the expressions of GRP78,ATF4 and CHOP were significantl
