摘要 基于微滴式数字聚合酶链式反应(droplet digital polymerase chain reaction,ddPCR)技术建立了定量分析马铃薯成分的检测方法。选取马铃薯的单拷贝光诱导组织特异性ST-LS1(light-inducible tissue-specific)基因作为目标基因,建立了马铃薯ST-LS1基因拷贝数浓度与马铃薯样品质量之间的线性关系;然后采用模拟样品验证该方法的准确性和适用性。结果表明:所设计的STLS1基因引物及探针具有良好的特异性,可对质量分数为5%及以上的马铃薯成分进行准确定量。此方法适用于市场监管中掺假鉴别及定量检测。 Based on the droplet digital polymerase chain reaction(ddPCR)technology,a method for quantitative of potato components was established.The single-copy ST-LS1 gene was selected as the target gene and a formula for calculating potato weight based on target gene copy number was established.Then use simulated samples to verify the accuracy and applicability of the method.The results showed that the designed primers and probe had good specificity and could accurately quantify potato components with a mass percentage of 5%or more.The method was suitable for adulteration identification and quantitative detection in market supervision.
