摘要
目的基于现代文献挖掘中医药治疗心脏X综合征的用药规律及核心药物,并探讨核心药物的作用机制。方法检索中国知网、维普、万方数据库中应用中药治疗心脏X综合征的文献,构建组方数据库,运用中医传承辅助平台软件(2.5版)挖掘心脏X综合征的用药规律及核心药物。使用BATMAN-TCM系统对核心药物进行疾病富集、KEGG通路分析,绘制靶点-通路-疾病网络可视图,探析核心药物治疗心脏X综合征的作用机制。结果通过筛选,共纳入符合条件的处方84首,涉及中药140味,药用频次>10次的中药有22味,排名前10位的中药分别为丹参、川芎、当归、甘草、瓜蒌、红花、枳壳、柴胡、茯苓、薤白。药物四气以温、寒、平为主,五味以苦、甘、辛为主。通过关联规则分析,得到处方中使用频次≥20次的药物组合7对,包含川芎、丹参、当归、红花、瓜蒌、甘草6味中药;得到药物组合规则6条,分别为:红花→丹参,红花→川芎,瓜蒌→丹参,川芎→丹参,当归→川芎,当归→丹参,挖掘出治疗心脏X综合征的核心药物为丹参、川芎、当归、瓜蒌、红花。KEGG通路分析发现这5味核心药物的富集疾病与心脏X综合征相关的有8种,潜在信号通路13条。结论临床治疗心脏X综合征药物多以活血药、理气药、化痰药为主,核心药物治疗心脏X综合征的作用机制可能与血管内皮生长因子、酪氨酸激酶受体、磷脂酰肌醇3-激酶/蛋白质丝氨酸苏氨酸激酶、钙离子、环鸟苷酸依赖的蛋白激酶、雌激素信号通路有关。
Objective Based on the modern literature,to explore the prescription rule and core drugs of traditional Chinese medicine in the treatment of heart X Syndrome and to explore the action mechanism of core drugs.Methods The literature on the application of traditional Chinese medicine in the treatment of heart X syndrome in CNKI,Weipu,and Wanfang databases were searched,the prescription database was constructed,and the TCM inheritance assistance platform software(version 2.5)was used to excavate the prescription rule and core drugs for heart X syndrome.The BATMANN-TCM system was used to conduct disease enrichment and KEGG pathway analysis of the core drugs,and the target-pathway-disease network was drawn to explore the action mechanism of core drugs in the treatment of heart X syndrome.Results A total of 84 qualified prescriptions were included,involving 140 Chinese medicines,22 with frequency of more than.The top 10 Chinese medicines were salvia miltiorrhiza,chuanxiong,angelica angelica,liquorice,fructus trichosanthis,safflower,fructus aurantii,bupleurum,poria cocos,stamen allium.Four properties were mostly warm,cold,flat;the five flavours were mostly bitter,sweet,pungent.Through association rule analysis,7 pairs of drug combinations with the use frequency≥20 in the prescription were obtained,including 6 traditional Chinese medicines:Chuanxiong,salvia miltiorrhiza,angelica sinensis,safflower,trichosanthis,and licorice root.Six drug combination rules were obtained,namely:safflower-Danshen,safflower-Chuanxiong,trichosanthis-Danshen,Chuanxiong-Danshen,angelica sinensis-Danshen.The core drugs for the treatment of heart X syndrome were found as follows:Danshen,Chuanxiong,angelica,trichosanthis,safflower.KEGG pathway analysis found that the enrichment of these 5 core drugs was associated with heart X syndrome in 8 kinds of diseases and 13 potential signaling pathways.Conclusion Most of the drugs used in the clinical treatment of heart X syndromeare blood activating drugs,qi regulating drugs,and phlegm-reducing drugs.The
