摘要
采用不断发展的基因编辑技术,近年来对猪的基因组改造不仅构建出涵盖心血管疾病、神经退行性疾病、新陈代谢疾病、癌症、免疫缺陷等各方面的人类疾病模型,而且通过对猪进行基因编辑实现了降低异种移植免疫排斥、敲除猪内源性逆转录病毒以及生产嵌合体动物等,使猪异种器官移植的应用更前进了一步。综述梳理基因编辑技术,尤其是CRISPR∕Cas9技术在异种器官移植及人疾病模型构建方面的研究进展。
With the usage of the recently developed genome editing technologies,extensive predetermined modifications have been incorporated into the pig genome,not only to build a variety of human disease models,covering cardiovascular disease,neurodegenerative disease,metabolic disease,cancer,immune deficiency and other aspects,but also to make the application of pig xenotransplantation a step further by reducing the immune rejection of xenotransplantation,knock-out of the porcine endogenous retroviruses(PERVs),as well as production of chimeric animals.In this review,we provide a brief introduction of pig genome editing and the related progress,especially for CRISPR∕Cas9 technology,with regard to the xenotransplantation and disease model areas.
