CD81、CCL26基因在黔北麻羊不同性腺组织中的表达研究 被引量:1

摘要 CD81和CCL26基因是影响哺乳动物性腺细胞融合的重要因子,但其在黔北麻羊性腺组织中的表达情况尚不清楚。为研究CD81和CCL26基因在黔北麻羊不同组织中的表达量,本实验以单、多羔黔北麻羊为研究对象,提取下丘脑、垂体、子宫、输卵管、卵巢组织的RNA,并将5种性腺组织RNA逆转录合成第一链cDNA,随后采用q-PCR技术检测CD81、CCL26基因的mRNA在单、多羔黔北麻羊不同性腺组织中的表达水平。结果表明:黔北麻羊性腺组织中CD81、CCL26基因的mRNA均有表达,2种基因均在单、多羔卵巢中的表达量最高;CD81基因在单羔组子宫中的表达量最低;CD81基因在多羔组、CCL26基因在单多羔组下丘脑中的表达量均最低;组间差异表达量分析可知,CD81基因在多羔组子宫的表达量显著高于单羔组;CCL26基因在单羔组卵巢和输卵管的表达量显著高于多羔组,在单羔组子宫的表达量极显著高于多羔组。本实验结果提示,CD81和CCL26基因可能与山羊繁殖能力相关,也为初步揭示山羊繁殖的分子调控机制提供了参考依据。 CD81 and CCL26 genes are important factors affecting mammalian gonadal cell fusion,but their expression in animal gonadal tissues is unclear.In order to study the expression levels of CD81 and CCL26 genes in different tissues of Qianbei Ma goat,single and multi-lamb Qianbei Ma goat were chose as the research object,and RNA was extracted from hypothalamus,pituitary,uterus,fallopian tube and ovarian tissue,and five gonad tissues were reverse transcribed into firststrand cDNA,and the expression levels of CD81 and CCL26 mRNA in different gonad tissues were detected by q-PCR.The results showed that the mRNAs of CD81 and CCL26 genes were expressed in gonads of Qianbei Ma goat,and both genes had the highest expression in single and multi-lamb ovary;CD81 gene had the lowest expression in uterus of single-lamb group;CD81 gene in multi-lamb group and the CCL26 in single-lamb group have the lowest expression in hypothalamus.Analysis of the differential expression between the groups shows that the expression of CD81 gene in uterus lamb group is significantly higher than that in single-lamb group;CCL26 is expressed in ovary and fallopian tubes.The expression level of single lambs was significantly higher than that of multiple lambs,and the expression of single lambs in uterus was significantly higher than that of multiple lambs.The results of this experiment suggest that the two genes CD81 and CCL26 may be related to goat reproductive ability,and also provide a reference for preliminary revealing the molecular regulation mechanism of goat reproduction.
