摘要
目的探讨在M-sol保存液中添加L-肉碱(LC)对体外保存血小板的影响。方法采集无偿献血志愿者的全血,采用白膜法制备浓缩血小板。将8袋ABO同型的浓缩血小板汇集,配制M-sol为血小板保存液。依据保存介质和LC浓度不同分为6组,即血浆组、M-sol组、M-sol+0.5mmol/L LC组、M-sol+5mmol/L LC组、M-sol+15mmol/L LC组和M-sol+25mmol/L LC组。置于(22±2)℃持续振荡保存,分别于保存d0、3、7取样。用全自动血细胞分析仪检测血小板计数(Plt)、平均血小板体积(MPV)、血小板分布宽度(PDW),用流式细胞仪检测血小板膜外磷脂酰丝氨酸(PS)和CD62p的阳性率,用半自动生化分析仪检测葡萄糖和乳酸含量,并计算保存7d期间葡萄糖消耗量和乳酸生成量。结果随保存时间的延长,各组Plt均下降,MPV和PDW均升高,在同一时间点各组间差异无统计学意义(P>0.05)。PS和CD62p阳性率逐渐升高,保存d7时,M-sol+15mmol/L LC组PS和CD62p阳性率低于其他各组(P<0.05)。葡萄糖消耗量和乳酸生成量M-sol+15mmol/L LC组最低(P<0.05)。结论将合适浓度的LC加入M-sol血小板保存液中,可降低血小板贮存损伤。
Objective To investigate the effect of L-carnitine(LC)added to M-sol preservation solution on platelet preservation in vitro.Methods Whole blood of voluntary blood donors was collected,and platelet concentrates were derived from buffy coat pools.8 bags of ABO-identical platelet concentrates were pooled and M-sol was prepared as platelet preservation solution.According to different preservation solution and LC concentration,they were divided into six groups:plasma group,M-sol group,M-sol+0.5 mmol/L LC group,M-sol+5 mmol/L LC group,M-sol+15 mmol/L LC group and M-sol+25 mmol/L LC group.They were stored at(22±2)℃with gentle agitation,and sampled on days 0,3 and 7 of storage,respectively.The platelet count(Plt),mean platelet volume(MPV),platelet distribution width(PDW)were detected by automatic blood cell analyzer,the positive rates of phosphatidylserine(PS)and CD62 p outside the platelet membrane were detected by flow cytometer,the contents of glucose and lactate were detected by semi-automatic biochemical analyzer,and the glucose consumption and lactate production during 7 days of storage were calculated.Results As the storage prolonged,Plt decreased,MPV and PDW increased in all groups,and few significant differences at the same time point were notable by groups(P>0.05).The positive rates of PS and CD62 p increased gradually.On the 7 th day of storage,the positive rates of PS and CD62 p in M-sol+15 mmol/L LC group were lower than those in other groups(P<0.05).Glucose consumption and lactate production were the lowest in M-sol+15 mmol/L LC group(P<0.05).Conclusion Adding appropriate concentration of LC into M-sol platelet preservation solution can reduce platelet storage lesion.
