摘要
目的检测低脂饮食治疗的T2DM患者不同阶段肠道菌群结构,分析其变化规律。方法选取2015年1月至2017年1月于舟山市普陀区人民医院内分泌科就诊的10例新诊断T2DM患者,给予长期低脂饮食,初诊及治疗1、3、6个月后分别采集粪便样本,分为D0、D1、D2、D3组。同期采集10名本院健康体检人群(NC组)粪便样本。所有样本采用高通量测序进行聚类、多样性分析及组间比较。结果两组肠道微生物在属水平上差异菌群较多。治疗过程中经菌群结构重组,D3组差异菌群相对丰度与NC组接近,菌群结构趋向于NC组。结论治疗不同阶段,微生物相对丰度不断变化。长期低脂饮食后,肠道菌群结构趋向正常。
Objective To detected the structure of intestinal microflora at different stages in patients with type 2 diabetes mellitus(T2DM)adhering to a low-fat diet,and to analyze the changes of intestinal microbes in patients with T2DM during the treatment.Methods A total of 10 newly diagnosed T2DM patients were selected from the Department of Endocrinology Putuo District People’s Hospital in Zhoushan City during January 2015 to January 2017.They were treated with long-term low-fat diet.Fecal samples were collected at baseline and after 1,3 and 6 months of treatment(D0,D1,D2,D3 group).Meanwhile,fecal samples were collected from 10 healthy subjects underwent health examination in our hospital(NC group).High throughput sequencing was used for clustering,diversity analysis and inter group comparison.Results At the genus level,there were many different floras between the two groups.During the treatment process,after the reorganization of the microflora structure,the relative abundance of differential floras and the structure of the flora in T2DM patients tended to those of healthy individuals.Conclusion The relative abundance of microorganisms was constantly changing at different stages of treatment.After a long-term low-fat diet,the intestinal flora tends to a healthy flora structure.
