甲磺酸伊马替尼增强5-氨基酮戊酸光动力疗法对皮肤鳞状细胞癌细胞杀伤作用探讨 被引量:4

摘要 目的探讨5-氨基酮戊酸(ALA)和/或甲磺酸伊马替尼对HaCaT角质形成细胞、皮肤鳞状细胞癌(cSCC)细胞A431内卟啉含量及三磷酸腺苷结合盒转运体G2(ABCG2)蛋白表达的影响,为进一步提高光动力的临床疗效提供理论依据。方法本研究采用表皮HaCaT角质形成细胞、cSCC细胞A431来源的人细胞系,在有或无甲磺酸伊马替尼的情况下,用ALA孵育这2种细胞。采用Western-Blotting方法测定ABCG2蛋白含量,酶联免疫吸附测定(ELISA)法检测细胞内卟啉物质的含量。结果在HaCaT细胞和A431细胞中,与单一使用ALA对A431细胞作用相比,ALA与甲磺酸伊马替尼联合作用可提高原卟啉物质在A431细胞内的蓄积作用,增强光动力疗法(PDT)诱导细胞死亡的作用。ALA与甲磺酸伊马替尼联合作用可显著降低ABCG2蛋白的表达,增强PDT治疗对A431细胞的杀伤作用。结论本研究揭示ALA-PDT治疗cSCC敏感性降低的可能原因,为采用ALA-PDT联合甲磺酸伊马替尼进一步改善PDT治疗的疗效提供理论基础。 Objective In order to investigate the effects of 5-aminolevulinic acid(ALA)and/or imatinibmesylate on the content of porphyrin and the expression of ABCG2 protein in HaCaT keratinocytes and skin squamous cell carcinoma cell A431,so as to provide a theoretical basis for further improving the clinical efficacy of photodynamics therapy(PDT).Methods In this study,human cell lines derived from epidermal HaCaT keratinocytes and skin squamous cell carcinoma cell line A431 were incubated with ALA with or without imatinib mesylate.Western blotting method was used to determine the content of adenosine triphosphate(ATP)binding cassette transporter G2(ABCG2),and enzyme-linked immunosorbent assay(ELISA)was used to detect the content of porphyrin in cells.Results In HaCaT cells and A431 cells,compared with the effects of ALA alone on A431 cells,the combination of ALA and imatinib mesylate could increase the accumulation of protoporphyrins in A431 cells,and enhance the PDT induced cell death.The combination of ALA and imatinib mesylate could significantly reduce the expression of ABCG2 protein and enhance the killing effects of PDT on A431 cells.Conclusion This study reveals the possible reasons for the decreased sensitivity of ALA-PDT in the treatments of skin squamous cell carcinoma,and provides theoretical basis for further improving the efficacy of PDT with ALA-PDT and imatinib mesylate.
