摘要
为了探究日粮中添加不同比例葡萄籽对多浪羊瘤胃真菌菌群结构的影响,选用15只安装永久瘤胃瘘管的健康多浪羊羯羊,将试验羊随机分成5组,分别饲喂添加0%(对照组)、4%(Ⅰ组)、8%(Ⅱ组)、12%(Ⅲ组)和16%(Ⅳ组)葡萄籽的全混合日粮。正式试验期的第10 d,于晨饲前采集瘤胃食糜,用高通量测序技术测定瘤胃液中真菌结构及其多样性。结果表明:(1)与对照组相比,Ⅳ组瘤胃真菌的菌群多样性增大(P<0.05)。(2)多浪羊瘤胃真菌门水平上的优势菌门为新丽鞭毛菌门(Neocallimastigomycot)和子囊菌门(Ascomycota)。Ⅱ组和Ⅳ组新丽鞭毛菌门的相对丰度较对照组分别降低了42.59%、48.24%(P>0.05),Ⅱ组子囊菌门的相对丰度较对照组增加了78.76%(P<0.05)。(3)真菌属水平上相对丰度较高的菌属为厌氧鞭菌属(Anaeromyces)、梨囊鞭菌属(Piromyces)、未注释粪壳菌纲(unclassifiedcSordariomycetes)、枝孢属(Cladosporium)、未注释肉坐菌目(unclassifiedoHypocreales)、Vishniacozyma、支顶孢菌属(Acremonium)、未注释真菌(unclassifiedkFungi)、阶菌属(Stephanonectria)、曲霉属(Aspergillus)。(4)Ⅰ组厌氧鞭菌属相对丰度最高为67.52%,较对照组增加34.42%(P<0.05);Ⅱ组梨囊鞭菌属相对丰度最低,较对照组降低79.68%(P>0.05);Ⅱ组Latorua相对丰度高于对照组(P<0.05)。综上,日粮中添加葡萄籽可增加瘤胃真菌菌群多样性,影响部分菌群相对丰度。
To investigate the effect of different proportions of grape seeds on the rumen fungi flora in Duolang sheep. 15 healthy Duolang sheep emasculated with ruminal fistula were randomly divided into 5 groups and fed 5 Diets with different proportions grape seeds of 0%(control group), 4%(group Ⅰ), 8%(group Ⅱ), 12%(group Ⅲ) and 16%(group Ⅳ). On the 10 th day of the formal trial period, rumen chyme was collected before morning feeding, and the structure and diversity of rumen fungi was measured by illumina miseq sequencing technology. The results showed that:(1) The diversity of rumen fungi in the group Ⅳ increased compared with the control group(P<0.05). The dominant phylum in the rumen fungi of Duolang sheep were Neocallimastigomycota, Ascomycota. The relative abundances of Neocallimastigomycota in the group Ⅱ and group Ⅳ were lower 42.59% and 48.24% of control group(P>0.05). The relative abundances of Ascomycota in the group Ⅱ were higher 78.76% of control group(P<0.05).(3) At the rumen fungal level of genus,the higher relative abundance at Duolang sheep were Anaeromyces, Piromyces, unclassified_c__Sordariomycetes, Cladosporiu, unclassified_o__ Hypocreales, Vishniacozyma, Acremonium, unclassified_k__Fungi, Stephanonectria, Aspergillus.(4) The relative abundance of Anaeromyces in the group Ⅰ was 67.52%, higher than 34.42% of control group(P<0.05). The relative abundance of Piromyces in the group Ⅱ was lower than 79.68% of control group(P>0.05). The relative abundance of Latorua in the group Ⅱ was higher than control group(P<0.05).In conclusion, the addition of grape seeds in the diet could increase the diversity of rumen fungi and affect the relative abundance of some of them.
