粪便中肠道病毒两种检测方法的应用比较

Comparison of Two Enterovirus-detecting Methods in the Stool

摘要 目的比较分析核酸检测和病毒分离这两种方法对健康儿童粪便中肠道病毒(enterovirus,EV)的检测结果。方法2019-08-27在凉山彝族自治州采集健康儿童粪便标本,采用实时荧光PCR法进行EV核酸检测。使用RD细胞(人横纹肌肉瘤细胞)和Hep-2细胞(人喉癌上皮细胞)对标本进行病毒分离,并运用SPSS 22.0软件对这两种方法的检测结果进行统计学分析,以P<0.05界定为差异显著。结果300份标本中,EV核酸检测与病毒分离法分别检出123例和106例阳性标本,阳性率分别是41.00%和35.33%;这两种方法的检测结果差异显著(P<0.001)。RD细胞与Hep-2细胞分别检出96例和26例阳性标本,阳性率分别是32.00%和8.67%;这两种细胞对病毒的检测结果具有显著差异(P<0.001)。结论EV核酸检测侧重于对肠道病毒进行初步筛查,而病毒分离侧重于获得病毒分离物。在实际运用中,可联合使用EV核酸检测法和病毒分离法以提高病毒分离率和准确率。 Objective To compare and analyze two methods(nucleic acid detection and virus isolation)used for detection of enterovirus(EV)in the stool of healthy children.Methods Stool specimens of healthy children were collected from Liangshan Yi Autonomous Prefecture on August 27,2019.Real-time fluorescent PCR was used for EV nucleic acid detection.RD cells(human rhabdomyosarcoma cells)and Hep-2 cells(human laryngocarcinoma epithelial cells)were used to isolate the enterovirus.Statistical analysis of the detection results obtained from these two methods was performed using SPSS 22.0 software with significant difference of P-value lower than 0.05.Results Among the 300 specimens,EV nucleic acid detection method and virus isolation method tested 123 and 106 positive specimens,respectively,with the positive rates of 41.00%and 35.33%,respectively;the rate of positive cases of the two detection methods was statistically significant(P<0.001).RD cells and Hep-2 cells detected 96 and 26 positive samples,respectively,and the positive rates were 32.00%and 8.67%,respectively.The detection results obtained from the two types of cells were significantly different(P<0.001).Conclusion EV nucleic acid detection focuses on the initial screening of enteroviruses,while virus isolation method dedicates to obtain pure virus.In practical applications,the EV nucleic acid detection method and the virus isolation method could be combined to improve the virus isolation rate and accuracy.
