子痫前期孕妇胎盘及血清中AT1-AA和VCAM-1表达水平变化及其与病情严重程度关系 被引量:7

Changes in expression levels of angiotensin II type 1 receptor-activating antibody and vascular cell adhesion molecules-1 in placenta and serum of pregnant women with preeclampsia and their association with severity of illness

摘要 目的探讨子痫前期孕妇胎盘及血清中血管细胞黏附因子-1(vascular cell adhesion factor-1,VCAM-1)和抗血管紧张素Ⅱ1型受体自身抗体(anti-angiotensin II type 1 receptor autoantibody,AT1-AA)表达水平,并探讨其与病情严重程度的关系。方法选取30例正常分娩孕妇(对照组)、30例子痫前期孕妇(子痫前期组)和30例重度子痫前期孕妇(重度子痫前期组)。采用酶联免疫吸附测定法(ELISA法)检测3组孕妇分娩前外周血清中VCAM-1和AT1-AA水平,采用Real-Time PCR技术检测3组胎盘组织中VCAM-1 mRNA和AT1-AA mRNA表达水平。结果 3组孕妇外周血中VCAM-1和AT1-AA、胎盘组织中VCAM-1 mRNA和AT1-AA mRNA表达水平差异均存在统计学意义(P<0.05),重度子痫前期组(血清VCAM-1、AT1-AA含量以及组织VCAM-1,AT1-AA1mRNA表达水平分别为[(39.8±13.5)μg/L、(148.7±12.7)ng/L、(0.67±0.15)、(0.59±0.14)]高于对照组[(42.7±5.5)μg/L、(59.8±6.7)ng/L,(0.20±0.04)、(0.18±0.05)]和子痫前期组[(99.2±8.7)μg/L、(113.7±10.8)ng/L,(0.35±0.09)、(0.33±0.10)],子痫前期组高于对照组,差异均有统计学意义(P<0.05)。重度子痫前期组和子痫前期组孕妇中,外周血VCAM-1和AT1-AA表达水平呈正相关(P<0.05),胎盘组织中VCAM-1 mRNA和AT1-AA mRNA表达水平呈正相关(P<0.05)。结论子痫前期孕妇胎盘及血清中VCAM-1和AT1-AA表达上调,且与子痫前期严重程度密切相关,子痫前期发生可能与AT1-AA诱导VCAM-1表达有关。
