妊娠高血压综合征患者血清肿瘤坏死因子与循环内皮祖细胞水平的相关性研究 被引量:3

Correlation study of TNF-α and circulating endothelial progenitor cell levels in patients with pregnancy induced hypertension syndrome

摘要 目的研究妊娠高血压综合征患者血清TNF-a与循环内皮祖细胞(EPC)水平的相关性。方法选择2014年9月~2015年8月在华北石油总医院住院的12例妊娠高血压综合征孕妇作为研究对象,选择同期人院的12例正常孕妇作为对照。检测所有孕妇的血清TNF-a和循环EPC数量。同时,分离外周血单个核细胞在体外进行诱导培养获得EPC,并对其增殖、黏附和迁移能力进行检测。结果妊娠高血压综合征孕妇血清TNF-a水平较正常孕妇明显升高(t=2.53,P〈0.01),而循环EPC数量则较正常孕妇显著降低(t=1.73,P〈0.05),并且妊娠高血压综合征孕妇血清TNF-a水平与循环EPC数量呈显著负相关(r=-0.73,P〈O.05)。妊娠高血压综合征孕妇循环EPC的体外增殖、黏附和迁移能力与正常孕妇比较,都存在一定程度的下调(均P〈0.05)。并且,妊娠高血压综合征孕妇循环EPC的体外增殖、黏附和迁移能力均与其血清TNF-a水平呈显著负相关(均P〈0.05)。结论妊娠高血压综合征的发生与血管内皮受损和炎性反应所导致的循环EPC数量和功能的下调存在明显的相关性。 Objective To assess the relativity of TNF-a and circulating endothelial progenitor cell (EPC) levels in the patients with pregnancy induced hypertension syndrome. Methods Twelve hospitalized patients with pregnancy induced hypertension syndrome from September 2014 to August 2015 in the General Hospital of Huabei Oil Field were selected as study objects, 12 normal pregnant women were selected as the control. The levels of plasma TNF-a and numbers of circulating EPCs of all pregnant women were assessed. And the peripheral blood mononuclear cells were separated and cultured in vitro, EPCs were obtained. The abilities of EPC proliferate, adhesion, migration were assessed. Results The levels of plasma TNF-a were increased significantly in the patient with pregnancy induced hypertension syndrome compared with normal pregnant women (t=2.53, P 〈 0.01), while the number of circulating EPC decreased (t=1.73, P 〈 0.05), and there was a significant negative correlation between the plasma levels of TNF-(x and the number of circulating EPC in patient with pregnancy induced hypertension syndrome (r=-0.73, P 〈 0.05). Meanwhile, the proliferation, adhesion and migration abilities of EPC in patients with pregnancy induced hypertension syndrome were all decreased compared with those normal pregnant women (all P 〈 0.05). Furthermore, there were significant negative correlation between the levels of plasma TNF-a and the functions of circulating EPC in patients with pregnancy induced hypertension syndrome (all P 〈 0.05). Conclusion The occurrence of pregnancy induced hypertension syndrome is correlated with the clown regulation of circulating endothelial progenitor number and functions evidently, which resulted from the vascular endothelium lesion and inflammatory reaction.
