健康/根腐病黄连根际土壤真菌组成与根部可培养真菌组成分析 被引量:3

摘要 【目的】揭示黄连根腐病与根部可培养真菌及根际土壤真菌组成的关系。【方法】用高通量测序法测定健康植株和根腐病植株根际土壤的真菌组成,并用组织分离法分离鉴定其根部真菌。【结果】病株和健株根际土壤真菌多样性、种属没有显著性差异。健株根和病株根中分离出的真菌组成有明显差别:病株根中优势种是Phoma herbarum;优势属是Phoma sp.、Ilyonectria sp.。健株根中优势种是DSE真菌Cadophora orchidicola;优势属是Cadophora sp.。黄连根部可培养真菌的Shannon-Wiener指数种水平上病株均比健株高,属水平上健株比病株高。【结论】土壤中真菌数量及组成对根腐病的发生没有显著影响;Ilyonectria sp.可能包含病原真菌。根部真菌属水平多样性的降低可能与根腐病发生相关。 【Objective】The present paper aimed to disclose the relationship between rhizosphere soil,root fungi composition and root rot disease of C.chinensis.【Method】The fungi composition of rhizosphere soil was analyzed by Illumina high-throughput Sequencing and the fungi of healthy and rot roots were isolated and identified.【Result】There was no significant difference between the fungi diversity,OTUs and abundance of healthy and root rot individual rhizosphere soil.There was obvious difference between the fungi composition of healthy and rot root.Phoma herbarum,Phoma sp.and Ilyonectria sp.were dominant in rot root while Cadophora orchidicola and Cadophora sp.were dominant in healthy one.The Shannon-Wiener index of cultivable fungi in rot root was higher than that of healthy one at species level,while lower at genus level.【Conclusion】The fungi composition of rhizosphere soil had no significant effect on root rot disease.Ilyonectria sp.may contain pathogen.The decrease of fungi diversity at genus level in C.chinensis root may have relation with root rot disease.
