摘要
为探究miRNA-1298-5p与TGF-βR1基因在内蒙古绒山羊绒毛生长周期中的差异表达对绒毛生长的影响,选取内蒙古优质品种阿尔巴斯白绒山羊绒毛生长的休止期、生长初期、生长旺盛期及退行期的体侧皮肤,提取总RNA,经DL2000紫外光度计和琼脂糖凝胶检测其纯度和浓度后,选取S-18S基因为内参,利用Primer 5设计相应的引物序列,通过实时荧光定量的方法对非编码miR-1298-5p与其潜在靶向基因TGF-βR1在绒毛不同生长时期皮肤的表达进行检测并通过△△Ct值法计算。结果表明:miR-1298-5p及TGF-βR1基因在阿尔巴斯白绒山羊皮肤4个不同发育时期均有表达;miR-1298-5p、TGF-βR1基因由绒毛生长初期到旺盛期表达量均增高,miR-1298-5p由旺盛期到退行期表达量增高,退行期到休止期降低;TGF-βR1基因由旺盛期到退行期表达量降低,退行期到休止期增高,二者表达量呈相反趋势,推测miR-1298-5p可能通过负向调控TGF-βR1基因在皮肤不同发育时期的表达,从而调控内蒙古绒山羊绒毛的生长。
In order to investigate the effect of differential expression of miRNA-1298-5p and TGF-βR1 gene in Cashmere growth cycle of cashmere goats,the skins were collected from the cashmere catagen;initial anagen;vigorous anagen and telogen of Arbas white cashmere goats in Inner Mongolia as the materials,the total RNA was extracted、purified and measured by DL2000 ultraviolet photometer and agarose gel.Then S-18S gene was selected as the internal reference and primer sequences were designed using Primer5.The expression of miR-1298-5p and its potential target gene TGF-βR1 at different growth stages of cashmere was tested by real-time fluorescence quantitative and calculated by △△Ct.The results showed that miR-1298-5p and TGF-βR1 gene were expressed at different skins in four different cashmere development periods of Inner Mongolia cashmere goats.The expression levels of miR-1298-5p and TGF-βR1 genes increased from initial anagen to vigorous anagen of cashmere growth,and the expression levels of miR-1298-5p increased from vigorous anagen to telogen and decreased from telogen to catagen.The expression level of TGF-βR1 gene decreased from vigorous anagen to telogen,and it increased drom teloge to catagen.The expression levels of the two showed the opposite trend.It is speculated that miR-1298-5p may negatively regulate the expression of TGF-βR1 gene at different developmental stages of the skin to regulate Inner Mongolian cashmere growth of cashmere goats.
