异丙酚通过EGFR/p38信号通路诱导胃癌细胞周期阻滞及相关机制研究 被引量:4

摘要 目的探讨异丙酚通过表皮生长因子受体(EGFR)/p38信号通路诱导胃癌细胞周期阻滞及相关机制。方法对数生长期的胃癌细胞系HGC-27分为3组,对照组、实验1组、实验2组给予无血清培养液、含1μg/mL异丙酚的无血清培养液、含10μg/mL异丙酚的无血清培养液处理6 h与12 h。采用四甲基偶氮唑盐(MTT)法检测细胞增殖,Transwell小室实验检测细胞侵袭,碘化丙啶(PI)染色法检测细胞周期,蛋白质印迹法检测蛋白表达。结果处理6 h与12 h后,实验1组与实验2组的细胞增殖、细胞侵袭指数、EGFR和p38蛋白相对表达水平低于对照组,实验2组也低于实验1组,差异均有统计学意义(P<0.05)。处理6 h与12 h后,实验2组与实验1组的S期细胞比率低于对照组,G2/M期细胞比率高于对照组,差异均有统计学意义(P<0.05),实验2组较实验1组细胞周期比率变化更显著,差异均有统计学意义(P<0.05)。结论异丙酚在胃癌细胞中的应用能阻滞细胞周期,抑制EGFR、p38蛋白的表达,从而抑制细胞增殖与侵袭,且具有剂量依赖性。 Objective To investigate the mechanism of propofol inducing cycle arrest of gastric cancer cells through epidermal growth factor receptor(EGFR)/p38 signaling pathway.Methods The gastric cancer cell line HGC-27 in the logarithmic growth phase were divided into three groups.The control group,experimental group 1 and experimental group 2 were given serum-free medium,serum-free medium containing 1μg/mL propofol,serum-free medium containing 10μg/mL propofol treatment for 6 h and 12 h.The MTT method were used to detect cell proliferation,the Transwell chamber experiment were used to detect cell invasion,the PI staining method were used to detect cell cycle,and the Western blot method were used to detect protein expression.Results After 6 h and 12 h treatment,the cell proliferation,cell invasion index,relative expression levels of EGFR and p38 protein of experimental group 1 and experimental group 2 were lower than those of the control group,and experimental group 2 were also lower than experimental group 1,and compared the differences were statistically significantly(P<0.05).After 6 h and 12 h treatment,the ratio of S phase cells in experiment group 2 and experiment group 1 were lower than the control group,and the ratio of G2/M phase cells were higher than that in the control group,compared the difference were statistically significant(P<0.05),compared with the difference between the experiment group 2 and experiment group 1 were also statistically significant(P<0.05).Conclusion The application of propofol in gastric cancer cells can block the cell cycle,inhibit the expression of EGFR and p38 protein,thereby inhibit cell proliferation and invasion,and there are dose-dependent.
