鼠神经生长因子联合瑞舒伐他汀治疗糖尿病周围神经病变大鼠的疗效及机制研究 被引量:7

Therapeutic effect and mechanism of rat nerve growth factor combined with rosuvastatin on diabetic peripheral neuropathy in rats

摘要 目的分析鼠神经生长因子联合瑞舒伐他汀治疗糖尿病周围神经病变大鼠的疗效及机制。方法经腹腔注射链脲佐菌素(STZ)建立糖尿病周围神经病变大鼠模型。随机留取8只大鼠作为正常对照组(C组,等剂量0.9%氯化钠溶液灌胃+腹腔注射);32只糖尿病大鼠随机分成糖尿病模型组(M组,同上)、鼠神经生长因子组[T1组,5 mL/kg 0.9%氯化钠溶液灌胃+腹腔注射12μg/(kg·d)鼠神经生长因子]、瑞舒他汀组[T2组,5 mg/(kg·d)瑞舒他汀灌胃+腹腔注射12μg/(kg·d)0.9%氯化钠溶液]和联合治疗组[T3组,5 mg/(kg·d)瑞舒他汀灌胃+腹腔注射12μg/(kg·d)鼠神经生长因子],均连续治疗12周。给药前、给药后4周、给药后12周检测各组大鼠空腹血糖;给药后4周、给药后12周检测各组大鼠总胆固醇含量;给药后12周检测各组大鼠肿瘤坏死因子α(TNF-α)、白细胞介素-6(IL-6)、白介素-10(IL-10)、坐骨神经传导速度(SNCV)检测,并取坐骨神经行血管内皮生长因子(VEGF)mRNA、微血管密度(MVD)及免疫组化结果。结果给药前、给药后4周、给药后12周,与C组相比,M组、T1组、T2组和T3组大鼠血糖含量均显著升高(P<0.05);给药后12周,3个治疗组大鼠血糖含量均较M组显著降低(P<0.05),3个治疗组相比,差异无统计学意义(P>0.05)。给药后4周,与C组相比,M组、T1组、T2组和T3组大鼠血液中总胆固醇含量均显著升高(P<0.05);给药后12周,与M组相比,T1组大鼠血液中总胆固醇含量差异无统计学意义(P>0.05),T2组和T3组大鼠血液中总胆固醇含量均显著降低(P<0.05)。给药后12周,与C组相比,M组大鼠血液中TNF-α、IL-6及IL-10表达量均显著升高,VEGF mRNA及VEGF、eNOS、nNOS蛋白表达量、SNCV、MVD均显著降低(P<0.05);与M组相比,T1组、T2组和T3组大鼠血液中TNF-α、IL-6表达量显著降低,IL-10、VEGF mRNA及VEGF、eNOS、nNOS蛋白表达量、SNCV、MVD均显著升高(P<0.05);与T1组和T2组相比,T3组大鼠血液中TNF-α� Objective To analyze therapeutic effect and mechanism of rat nerve growth factor combined with rosuvastatin on diabetic peripheral neuropathy in rats.Methods A rat model of diabetic peripheral neuropathy was established by intraperitoneal injection of streptozotocin.8 rats were randomly selected as the normal control group(group C,the same dose of 0.9%sodium chloride solution intragastrically+intraperitoneal injection),and 32 diabetic rats were randomly divided into diabetes model group(group M,same as above),rat nerve growth factor group(group T1,5 mL/kg 0.9%sodium chloride solution intragastric administration+intraperitoneal injection of 12μg/(kg·d)rat nerve growth factor),rosustatin group[group T2,5 mg/(kg·d)Rosustatin gavage+intraperitoneal injection of 12μg/(kg·d)0.9%sodium chloride solution]and the combined treatment group[group T3,5 mg/(kg·d)Rosustatin gavage+intraperitoneal injection of 12μg/(kg·d)mouse nerve growth factor],all groups were treated continuously for 12 weeks.The fasting blood glucose of the rats in each group was measured before,4 weeks and 12 weeks after the administration;the total cholesterol content of the rats in each group was measured at 4 weeks and 12 weeks after the administration;12 weeks after the administration,tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),interleukin-10(IL-10),sciatic nerve conduction velocity(SNCV)were detected in each group,and the sciatic nerve was taken for vascular endothelial growth factor(VEGF)mRNA,microvessel density(MVD)and immunohistochemical results.Results Before administration,4 weeks after administration,and 12 weeks after administration,compared with group C,the blood glucose levels of rats in group M,T1,T2 and T3 all increased significantly(P<0.05);after administration At 12 weeks,the blood glucose levels of rats in the 3 treatment groups were significantly lower than those in the M group(P<0.05),and there was no statistically significant difference between the 3 treatment groups(P>0.05).After 4 weeks of administration,compared wi
