摘要
目的:将强迫游泳应激作为刺激点应用于经前期综合征(premenstrual syndrome,PMS)肝气郁证大鼠模型制备,探究其与束缚法所得PMS大鼠模型之间行为学差异。方法:筛选动情周期规律、旷场评分相近大鼠纳入实验,随机分为空白组、模1组(束缚造模)、模1给药组(束缚造模+给药氟西汀)、模2组(改良造模法)、模2给药组(改良造模法+给药氟西汀)。给药组造模同时给予氟西汀(2.5 mg·kg-1)灌胃,空白组给予等量蒸馏水,共21 d。于每日早晚观测动情期变化,期间观测大鼠一般行为变化。造模结束进行旷场实验、糖水偏嗜实验、甩尾潜伏期测定,并持续观测动情周期。取大鼠卵巢称质量并计算脏器系数,病理切片观察大鼠卵泡发育情况及数目。结果:与空白组比较,模型组大鼠各项指标均出现异常,模2组异常程度较模1组更为严重,而给药组各项指标均有不同程度改善。本实验束缚法所得模型成功率为40%,改良造模法可使该模型成模率提升至60%。结论:强迫游泳刺激对大鼠一般行为变化、动情周期、旷场总路程和平均速度及静止时间、糖水敏感程度、甩尾潜伏期、卵巢比重及卵泡数目均有不同程度的影响,提示在原造模基础上加入冷热水交替强迫游泳刺激可复制经前期综合征模型,提高造模成功率。
Objective:To apply forced swimming stress as a stimulus point to prepare a rat model with premenstrual Liver Qi Depression Syndrome,and to explore the behavioral differences between it and the premenstrual syndrome(PMS)rat model obtained by restraint method.Methods:The rats with estrous cycle regularity and similar open field scores were selected into the experiment,and they were randomly divided into blank group,model 1 group(binding model),model 1 administration group(binding model+administration of fluoxetine),and model 2 group(improved modeling method),mode 2 administration group(improved modeling method+administration of fluoxetine).The administration group was given fluoxetine(2.5 mg·kg-1)by gavage at the same time for modeling,and the blank group was given the same amount of distilled water for 21 days.Observe the changes in the estrus period every morning and evening,and observe the general behavioral changes of the rats during the period.At the end of the model building,an open field experiment,a sugar water preference experiment,and a tail-flick latency measurement were performed,and the estrus cycle was continued to be observed in the next week.Take the rat ovary and weigh it,calculate the organ coefficient,and observe the development and number of rat follicles by pathological section.Results:Compared with the blank group,the rats in the model group were abnormal in all indicators,and the abnormality of the model 2 group was more serious than that of the model 1.The various indicators of the drug group improved to varying degrees.The success rate of the model obtained by the restraint method in this experiment is 40%,and the improved modeling method can increase the model formation rate to 60%.Conclusion:Forced swimming stimulation has different effects on the general behavioral changes,estrus cycle,total distance in the open field,average speed and resting time,sugar water sensitivity,tail-flick latency,ovarian specific gravity and number of follicles to varying degrees,suggesting that it is based o
