摘要
目的通过对一起新型冠状病毒引起的典型聚集性病例样本的核酸及抗体检测结果进行分析,为实验室优化新型冠状病毒检测策略提供参考。方法对一起新型冠状病毒引起的典型聚集性病例(共计5例阳性病例)的核酸检测及血清抗体检测结果,从样本类型、采集效率、阳性结果的出现和转阴的时间等方面进行比较和分析。结果不同病例各种样本排毒情况不尽相同。14份痰液样本和8份咽拭子样本内标基因(人类细胞管家基因)CT值采用校正t检验,t′=-2.149(校正自由度v=17.226),P<0.05,结果差异有统计学意义。胶体金方法血清抗体检测到阳性结果时间晚于呼吸道样本核酸检测到阳性结果的时间。结论呼吸道样本应尽量采集到痰液,应选用带内标试剂对呼吸道样本的采样效率进行评估;对出院或解除隔离的病例应同时采集呼吸道和肠道样本;胶体金试剂检测抗体在疾病早期诊断中意义不大。
Objective To analyze the results of nucleic acid and antibody detection of a typical clustering event,so as to provide a reference for optimizing the SARS-CoV-2 detection strategy in laboratory.Methods The results of nucleic acid test and serum antibody test of a typical clustering event(including 5 positive cases)caused by SARS-CoV-2 were compared and analyzed from the aspects of sample type,collection efficiency,appearance of positive results and the time of turning negative.Results The detoxification situation of different types of specimens of the cases was different.The CT values of the internal standard gene(housekeeper gene in human cells)in 14 sputum samples and 8 throat swabs samples were tested by corrected t-test,t′=-2.149(corrected freedom v=17.226),P<0.05.The differences were statistically significant.The time of getting positive results of serum antibody by colloidal gold method was later than that of respiratory tract samples by nucleic acid detection.Conclusion Sputum samples should be collected if possible,and the sampling efficiency of respiratory tract samples should be evaluated by the reagents with internal markers.The respiratory and intestinal specimens should be collected simultaneously from patients discharged from hospital or released from isolation.The detection of antibody by colloidal gold method is of little significance in early diagnosis.
