摘要
目的:采用网络药理学方法,筛选天麻-钩藤药对治疗帕金森病(PD)的关键靶点及其作用机制。方法:通过TCMSP、TCMID、NPASS、ETCM数据库挖掘其有效成分及关键靶点。经DisGeNET数据库收集PD相关基因。采用Cytoscape软件绘制成分-靶点-疾病可视化网络图。利用STRING数据库进行构建蛋白互作网络图,并进一步对关键靶点进行GO功能富集分析和KEGG通路富集分析。结果:天麻-钩藤药对中对PD有治疗作用的58种有效成分,作用于丝氨基酸/苏氨基酸激酶1(AKT1)、胱冬酶3(CASP3)、丝裂原活化蛋白激酶8(MAPK8)、白细胞介素-6(IL-6)、雌激素受体1(ESR1)等81个靶点。KEGG富集分析结果显示主要涉及到细胞凋亡、自噬、炎症等相关信号通路。结论:天麻-钩藤药对治疗PD具有多靶点、多途径的特点。对关键靶点及其相关通路的初步分析显示,本病与病毒性脑炎、动脉粥样硬化、癌症具有密切的联系,为今后PD病因、发病机制的研究和临床药物的开发提供了思路。
Objective:To obtain and analyze the key targets and molecular mechanism of Tianma(Gastrodiae Rhizoma)-Gouteng(Uncariae Ramulus Cum Uncis)herbal pair in the treatment of Parkinson’s disease(PD)by network pharmacology method.Methods:The main components and effective targets of Tianma-Gouteng were summarized through data mining of TCMSP,TCMID,NPASS,ETCM and PD-related genes were extracted from DisGeNET.The component-target-disease visual network was established by using Cytoscape software and PPI network was established by STRING database.KEGG pathway enrichment analysis and GO enrichment analysis were performed for key target proteins.Results:A total of 58 components were effective in treating Parkinson’s disease,which acted on 81 targets such as AKT1,CASP3,MAPK8,IL6 and ESR1.The KEGG pathway enrichment analysis showed autophagy,inflammation,apoptosis and other related signal pathways were involved.Conclusions:Tianma-Gouteng herbal pair has the characteristics of multi-pathway and multi-target action in the treatment of Parkinson’s disease.The study preliminarily analyzes the key targets and related pathways of Tianma-Gouteng,and finds that Parkinson’s disease is closely related to atherosclerosis,cancer and viral encephalitis,which provides ideas for the future study on the etiology and pathogenesis of Parkinson’s disease and the development of clinical drugs.
