摘要
目的:评价含附子-半夏的温阳化痰汤对多囊卵巢综合征(PCOS)大鼠的毒性。方法:以1 mg·kg-1剂量来曲唑溶液连续灌胃SD雌性大鼠21 d并建立PCOS大鼠模型,分别对正常大鼠给予温阳化痰汤干预、对PCOS模型大鼠给予温阳化痰汤和附子-半夏合煎液干预,给药4周后检测各组大鼠血清中肌酸激酶同工酶MB(CK-MB)、乳酸脱氢酶(LDH)、谷丙转氨酶(ALT)、谷草转氨酶(AST)、尿素氮(BUN)、肌酐(Cr)水平,并观察主要脏器心脏、肝脏和肾脏的形态学变化。结果:血液生化指标的检测结果与组织病理学结果基本一致PCOS模型大鼠给药附子-半夏后各生化指标水平显著升高,主要脏器损伤严重;PCOS模型大鼠给药温阳化痰汤后各生化指标水平降低,主要脏器的病理改变不明显;正常大鼠给药温阳化痰汤后各生化指标水平升高,对主要脏器可造成明显损伤,特别是对心肌细胞和肝细胞。结论:附子-半夏反药组合在温阳化痰汤中经配伍后其毒性会降低,且温阳化痰汤的毒性在生理和病理状态下的动物中存在较大差异,其对正常大鼠的毒性明显高于对PCOS大鼠。
Objective:To evaluate the toxicity of Wenyang Huatan Decoction with Aconiti Lateralis Radix Praeparata-Pinelliae Rhizoma in polycystic ovary syndrome(PCOS)rats.Methods:PCOS rat model was established by continuously administered SD female rats with letrozole solution at a dose of 1 mg·kg-1 for 21 days.Normal rats were intragastrically administered Wenyang Huatan Decoction.PCOS rats were intragastrically administered Wenyang Huatan Decoction or Aconiti Lateralis Radix Praeparata-Pinelliae Rhizoma decoction respectively.4 weeks after administration,blood biochemical indicators CK-MB,LDH,ALT,AST,BUN and CR of rats in each group were detected,and the morphological changes of the main organs,heart,liver and kidney were observed.Results:The results of blood biochemical indicators were basically consistent with those of histopathology.Compared with the model group,the serum levels of the six enzymes above were significantly increased in the Aconiti Lateralis Radix Praeparata-Pinelliae Rhizoma PCOS group,and the main organs were seriously damaged.Compared with the model group,the serum levels of six enzymes were decreased in the Wenyang Huatan Decoction PCOS group,and the pathological changes of the main organs were not obvious.Compared with the normal group,the serum levels of six enzymes were increased in the Wenyang Huatan Decoction normal group,the main organs were significantly damaged,especially the cardiomyocytes and hepatocytes.Conclusion:The toxicity of Aconiti Lateralis Radix Praeparata-Pinelliae Rhizoma in Wenyang Huatan Decoction was reduced after compatibility.The toxicity of Wenyang Huatan Decoction was significantly different in physiological and pathological animals,and its toxicity to normal rats was significantly higher than that to PCOS rats.
