板连败毒口服液的急性毒性与长期毒性试验 被引量:4

摘要 探索板连败毒口服液的急性毒性与长期毒性。急性毒性试验中,SPF级昆明小鼠预试验高剂量组灌胃达到6000 mg/kg,实验动物死亡为0/4,故将动物数量增加至10只(雌、雄各半),并且重复2次,观察实验动物中毒表现和死亡情况。长期毒性试验中,SPF级SD大鼠96只分为4组,每组24只(雌雄各半),空白对照组、板连败毒口服液高、中和低剂量组,给药期限30 d,停药后观察15 d。每天观察大鼠外观体征、饮水和食欲情况,每周称量体重,第30天和第45天分别处死16只和剩余部分,采血检测血常规、血液生化和解剖观察主要脏器病理学变化。急性毒性试验中,板连败毒口服液LD50大于6000 mg/kg,实际无毒。长期毒性试验中,体重增长趋势组间均无显著差异;组间血常规和血液生化指标均在正常范围内,其中红细胞数量等指标有波动,但其差异水平停药后恢复正常;重要器官的脏器系数及病理变化组间均无差异显著性。研究结果表明,板连败毒口服液实际无毒,在临床拟用量范围内使用安全。 The aim of this study was to explore the acute toxicity and long-term toxicity tests of Ban-lian Bai-du Oral-liquid.In acute toxicity test,the dosage in high dose group of pre-test mice reached 6000 mg/kg,and the death of experimental animals was 0/4.Therefore,the number of animals was increased to 10 per group the experiment was repeated twice to observe experimental animal poisoning performance and the mortality.In long-term toxicity test,ninety six SD rats were divided into 4 groups,including the normal group,Ban-lian Bai-du Oral-liquid high,middle and low dose groups,each group with 24 rats(half male and half female).30 days after administration,the rats were observed for 15 days after stopping drug.The appearance signs,drinking water and appetite of the rats were observed daily,and the weight was weighed weekly.Blood and organs were taken on the 30th and 45th days to detect blood biochemistry,blood routine parameters and pathological lesions of major organs.The acute toxicity test showed that the LD50 of Ban-lian Bai-du Oral-liquid was more than 6000 mg/kg,and there was no toxicity.The long-term toxicity test showed that the body weight gains were no significant differences.The blood routine and blood biochemical indexes in each group rats were within the normal range,and there were no differences in the main organ indicators.Among them,RBC fluctuated up and down,but the difference level in the 15-day recovery experiment gradually progressed to normal.Ban-lian Bai-du Oral-liquid is practically non-toxic,and it is safe to use in the range of clinical doses.
