摘要
目的:研究桑杏护肺颗粒(SXHFKL)对放射性肺损伤大鼠Th1/Th2免疫失衡的调节作用。方法:SD大鼠随机分为空白组(CON)、模型组(RILI)、强的松组(阳性对照,PDN 3.125mg/kg)、SXHFKL治疗组(高剂量:SH20mg/kg、中剂量:SM10mg/kg、低剂量:SL5mg/kg)。照射后第4w起,每4w各组随机抽取8只大鼠,ELISA法检测Th1相关细胞因子IL-12、IFN-γ及Th2相关细胞因子IL-4、IL-5、IL-13表达改变情况并采用RT-PCR法检测其mRNA丰度变化情况。FCM检测Th1/Th2细胞比值变化情况。结果:放疗12w后可引起Th1相关因子表达及mRNA丰度下调,Th2相关因子表达及mRNA丰度上调,Th1/Th2细胞比值下降。SXHFKL治疗各组于20wTh1/Th2细胞免疫失衡情况有所缓解,且与PDN无显著差异,但其各剂量组间无明显差异。结论:改善Th1/Th2细胞免疫失衡可能是ST保护肺损伤的作用机制之一。
Objective:Studying the regulatory effect of Sangxing Hufei Granule to the imbalance between Th1 and Th2 cells,which was one possible mechanism that Sangxing Hufei Granule used to protect lung tissue from radiation pulmonary injury.Methods:SD rats(male,200~250g)were randomly divided into normal group,model group,positive medicine group(prednisone group,3.125mg/kg),Sangxing Hufei Granule group(20g/kg/d,10g/kg/d,5gkg/d,radiation amount:14GY,respectively).All rats,excluding normal group,were given X ray chest single irradiation to building radiation pulmonary injury model.At the designed time(4th,8th,12nd,16th,20th,24th,28th,32nd weeks after irradiation),ELISA methods were used to assessed the mRNA level of the inflammatory factors,including Th1 relating factors IL-12,IFN-γand Th2 relating factors IL-4,IL-5,IL-13.Meanwhile,the expression of these inflammatory factors was detected by ELISA and the flow cytometry(FCM)assay was employed to evaluate the ratio of Th1 and Th2 cells in lung tissue.Results:The reduce level of Th1 relating factors and increasing level of the Th2 relating factors have been detected at 12 week.The decreasing radio of Th1 and Th2 cells also been detected in our studies which induced by radiation.Fortunately,Sangxing Hufei Granule could reverse these pathologic changes.However,the regulatory effect of Sangxing Hufei Granule to the imbalance between Th1 and Th2 cells was nondifference to prednisone,but the regulatory effect prednisone could be detected at 12 week and the similar effect of Sangxing Hufei Granule was observed at 20 weeks.It was strange that the regulatory effect of Sangxing Hufei Granule not showed dose-dependent fashion.Conclusion:The regulatory effect of Sangxing Hufei Granule to the imbalance between Th1 and Th2 cells would be one possible mechanism that Sangxing Hufei Granule is used to protect lung tissue from radiation injury.
